榄香烯联合热疗体外对肝癌SMMU7721细胞增殖的影响

The inhibitory effect of SMMC7721 Cell treated by Elemene in combination with hyperthermia

目的观察榄香烯联合热疗对肝癌细胞增殖的影响。方法采用肝癌细胞系SMMC7721体外培养,MTT法测定单纯热疗、单纯榄香烯及榄香烯联合热疗对细胞增殖的影响。另外用流式细胞术的方法测定对细胞增殖核抗原(PCNA)和细胞周期的影响。结果榄香烯联合热疗对肝癌细胞增殖的抑制作用比单纯热疗和单纯榄香烯强,并且能够降低细胞PCNA的表达,使细胞周期中的S细胞减少,阻滞细胞于G0/G1。结论 榄香烯联合热疗可以增强对肝癌细胞增殖的抑制作用。

Objective To investigate the inhibitory effects of Elemene combined with hyperthermia on human hepatoma carcinoma cell.Methods SMMC7721 cells were cultivated in vitro and treated with hyperthermia,Elemene,and Elemene in combined with hyperthermia.The inhibition rates were observed by MTT.The proliferating cell nuclear antigen（PCNA） expression and cell cycle were detected by flow cytometry.Results The inhibitory rate of the Elemene plus hyperthermia was higher than that of hyperthermia,Elemene.In addition,Elemene plus hyperthermia could significantly inhibit the growth of SMMC7721 cells and arrest cells in G0/G1 phase.Also the expression of PCNA protein was inhibited.Conclusion The Elemene combined with hyperthermia was effective on the inhibition of proliferation of hepatoma carcinoma cell.
