摘要
目的利用响应曲面法对金钱草总黄酮纯化工艺进行优化。方法选择跨膜压力、药液温度及药液浓度为自变量,总黄酮纯度为响应值,采用Box-Behnken设计研究各自变量及其交互作用对总黄酮提取率的影响。利用Design Expert软件对数据进行回归分析,得到回归方程的预测模型。结果金钱草总黄酮超滤的优化工艺为:在跨膜压力0.27 MPa、药液温度34℃、药液浓度0.24 g(生药)/ml的条件下进行错流循环超滤,总黄酮纯度的理论值为18.38%,实际值为18.20%。结论响应曲面法确定的工艺条件纯化金钱草总黄酮效果良好。
Objective To investigate optimum conditions for the ultrafiltration of total flavonoids from christina loosestrife herb by using response surface methodology.Methods Trans-membrane pressure,liquid temperature and the drug concentration were selected as independent variables,and the purity of total flavonoids was regarded as response value.All variables and their interactive effects on the purity of total flavonoids were also investigated with Box-Behnken Design.The predictive model of polynomial quadratic equation was analyzed by Design Expert software.Results The results indicated that the optimum extraction conditions of total flavonoids were as follows: the trans-membrane pressure 0.27 MPa,liquid temperature 34℃,the ratio of material to water 0.24 g/ml.Under the optimized condition,the predicted purity of the total flavonoids was 18.38%,and the actual value was 18.20%.Conclusion Optimum conditions are suitable for the ultrafiltration of total flavonoids from christina loosestrife herb by using response surface methodology.
