摘要
目的探讨外源性5’-核苷酸对大鼠酒精性肝损伤的影响。方法 60只雄性Wistar大鼠随机分成5组:空白对照组,酒精模型组,无核苷酸干预组,低、高剂量干预组。除空白对照组灌胃蒸馏水外,各组用50%酒精灌胃4w。处死空白对照组和酒精模型组大鼠,测定血清谷草转氨酶(AST)、谷丙转氨酶(ALT)、总甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、白蛋白(ALB)、总蛋白(TP)、超氧化物歧化酶(SOD)和丙二醛(MDA)水平,观察肝脏病理改变;剩余3组大鼠分别给予无核苷酸饲料和添加低、高剂量5’-核苷酸饲料干预喂养2w,观察干预效果。结果酒精灌胃4w后大鼠肝体比(A/G)升高,血清ALT、AST、TC、HDL-C水平升高,肝组织出现轻微的病理改变,造模成功;无核苷酸饲料干预后,各异常指标都趋于正常,大鼠表现出良好的自我恢复能力;而5’-核苷酸饲料干预后,大鼠血清TG水平降低,ALB、A/G水平升高,SOD活力增加。结论添加外源性5’-核苷酸能够促进大鼠白蛋白合成,调节脂质代谢,增强其抗氧化能力,有助于肝组织损伤后修复。
Objective To investigate the effects of exogenous 5'-nucleotides supplemented diets on early alcoholic liver disease in Wistar rats.Method To build alcoholic liver disease model,48 male Wistar rats were gavaged with 50% ethanol and 12 rats with distilled water as control.After 4 w,11 model rats died.Eight of the model rats and all control rats were killed to observe whether the model was established.Blood and hepatic tissue of the rats were collected.The level of alanine amino transferase(ALT),aspartate amino transferase(AST),triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C),total protein(TP),albumin(ALB),the activities of superoxide dismutase(SOD),and the content of malondialdehyde(MDA) in plasma were measured.Liver histology was assessed.In the later 2 w,the left 29 model rats were divided into 3 groups,9 rats were fed diets without nucleotides(MN),10 in each,with low and high dose of 5'-nucleotides(ML and MH) supplemented diets.Results Compared with controls,the model group had higher hepatosomatic index,higher plasma ALT,AST,TC,HDL-C level and liver histological study showed mild injury.The changes recovered 2w after alcohol intake.Furthermore,plasma TG was lower and SOD activities was higher in ML group,while plasma TG was lower and ALB,albumin/globulin,and SOD activities was higher in MI group than MN group.Conclusion Exogenous nucleotides from diet could promote restoration of alcoholic liver injury in rats.
