大蒜多糖及其硒化产物抗氧化活性的比较研究 被引量:8

STUDY ON ANTIOXIDATIVE ACTIVITIES OF GARLIC POLYSACCHARIDE AND SELENO-GARLIC POLYSACCHARIDE

摘要 目的比较大蒜多糖与硒化大蒜多糖对经过氧化氢(H2O2)诱导损伤的大鼠嗜铬瘤细胞株(PC12)的保护作用。方法用H2O2建立PC12细胞损伤模型,应用四氮唑蓝(MTT)比色法检测PC12细胞的活力;化学比色法测定细胞培养液中乳酸脱氢酶(LDH)释放量、细胞内和细胞培养液中丙二醛(MDA)含量及超氧化物歧化酶(SOD)的活性。结果硒化大蒜多糖、大蒜多糖500μg/ml及以上剂量组和亚硒酸钠12.50μg/ml及以上剂量组均能有效抑制由H2O2引起的细胞存活率的下降(P<0.01)。硒化大蒜多糖、大蒜多糖2000μg/ml可显著降低LDH释放量和细胞培养液及细胞内的MDA含量,以及提高SOD活性(P<0.01),表现出良好的抗氧化活性,而硒化大蒜多糖的抗氧化作用更为明显(P<0.05)。结论硒化大蒜多糖对经H2O2诱导损伤的PC12细胞具有保护作用,这种保护作用明显优于大蒜多糖或单纯硒,其作用机制可能与提高PC12细胞的抗氧化能力有关。 Objective To compare the protective effect between garlic polysaccharide and seleno-garlic polysaccharide on pheochromocytoma cells(PC12) injured by hydrogen peroxide(H2O2).Method An injured cell model of PC12 by H2O2 was stablished firstly.Cell viability was tested by MTT assay.Lactate dehydrogenase(LDH),malondialdelyde(MDA) and superoxide dismutase(SOD) in the culture medium and MDA,SOD in cells were measured by chemical colorimetry.Results Seleno-garlic polysaccharide and garlic polysaccharide at 500 ?g/ml and Na2SeO3 at 12.50 ?g/ml could resist the decrease in cell viability injured by H2O2(P〈0.01).Seleno-garlic polysaccharide and garlic polysaccharide at 2000 ?g/ml could decrease the release of LDH and MDA,and enhance the activity of SOD significantly(P〈0.01).They exhibited high antioxidant effect,more significantly increased by seleno-garlic polysaccharide than by garlic polysaccharide(P〈0.05).Conclusion Seleno-garlic polysaccharide has protective effect on PC12 injured by H2O2,better than garlic polysaccharide and sucrose-nano-Se.It may be related to increase of the antioxidant abilities of PC12.
