摘要
目的:观察活血通络凝胶对兔膝骨性关节炎血清超氧化物歧化酶(SOD)、过氧化脂(LPO)、软骨含水量、己糖醛酸和羟脯氨酸水平的影响。方法:将75日龄日本大耳家兔60只,随机分为空白组、模型组、对照组、治疗组,其中模型组30只,其余每组10只。除空白组外,其余各组家兔均采用改良的Hulth法造模。于造模后1周开始进行治疗,模型组给予关节腔内注射生理盐水0.2mL/次;对照组给予关节腔内注射透明质酸钠0.2mL/次;治疗组给予活血通络凝胶局部涂抹,0.1g/次,覆盖患膝为度,每天1次。4周为1疗程,共3疗程。疗程结束后,检测兔血清SOD、LPO以及患膝软骨含水量、己糖醛酸和羟脯氨酸含量。结果:治疗组血清SOD含量升高,与模型组比较,差异有显著性意义(P<0.05)。治疗组血清LPO含量明显降低,与模型组比较,差异有非常显著性意义(P<0.01);与对照组比较,差异有显著性意义(P<0.05)。治疗组软骨含水量明显降低,与模型组比较,差异有显著性意义(P<0.05)。治疗组己糖醛酸含量明显升高,与模型组、对照组比较,差异均有显著性意义(P<0.05)。治疗组羟脯氨酸含量明显升高,与模型组、对照组比较,差异均有显著性意义(P<0.05)。结论:活血通络凝胶可以有效改善膝骨性关节炎兔患膝氧自由基以及软骨生化,提高抗氧自由基的作用,减少氧自由基对膝关节软骨的损伤。
Objective: To observe the influence of Huoxue Tongluo Gel on serum superoxide dismutase(SOD), lipid peroxide (LPO), hexuronic acid and hydroxyproline levels, and cartilage water content of experimental knee osteoarthritis(KOA)in rabbits. Methods. Sixty 75-day-old Japanese white rabbits were divided into four groups randomly: normal group, model group, treatment group and control group. Thirty rabbits were in the model group, and 10 were in each of the other groups. The model was established by the modified Hulth method. The model group was treated with intraarticular injection of normal saline, 0.2 mL each time. The control group was treated with intraarticular injection of sodium hyaluronate, 0.2 mL each time. The treatment group was applied with Huoxue Tongluo Gel externally for the affected knee, 0.1g each time. Four weeks constituted one treatment course, and the treatment lasted 3 courses. After treatment, serum levels of SOD, LPO, hexuronic acid and hydroxyproline, and cartilage water content were detected. Results: After treatment, SOD level was increased (P 〈 0.05), LPO level was decreased (P 〈 0.05) and cartilage water content was reduced (P 〈 0.01) in the treatment group compared with those in the model group. The difference of LPO level between the treatment group and the control was also significant (P 〈 0.05). Hexuronic acid and hydroxyproline levels were both increased in the treatment group (P 〈 0.05 compared with those in the model group and the control group). Conclusion; Huoxue Tongluo Gel is effective on postponing degenerative changes of anthrodial cartilage through inhibiting the biological effect of oxygen free radical and promoting the biological effect of SOD.
