摘要
目的分析猪带绦虫成虫烯醇酶(enolase,Ts ENO)基因结构并预测其编码蛋白的结构和功能。方法利用生物信息网站如美国国家生物技术信息中心(NCBI)和瑞士生物信息学研究所的蛋白分析专家系统(ExPASY)中生物信息学分析工具,并结合其它分析软件,从获得猪带绦虫成虫全长cDNA质粒文库的表达序列标签(expression sequence tag,EST)中识别Ts ENO基因,分析该基因的结构并预测其编码的蛋白质的结构和功能特征。结果该基因与亚洲带绦虫烯醇酶基因的一致性为98%,相似性为99%;全长1 725bp,编码区为202~1162bp,编码320个氨基酸,为全长基因。无各种亚细胞定位序列,预测该蛋白为跨膜蛋白,与吸虫属的进化关系较近。结论从猪带绦虫成虫cDNA文库中筛选出烯醇酶基因,预测为跨膜蛋白,可能具有较好的免疫诊断抗原应用前景。
Objective To analyze and predict the structure and characteristics of enolase by bioinformatics in order to guide the experimental research for its biological function and application.Methods By using the analyzing tools of bioinformatics recognize enolase from the Taenia solium full-length cDNA plasmid libratory and analyze the gene structure,so as to analyze and predict physical-chemical characteristics,topological structure.Results The full gene was 1 725 bp in length,its coding region was 202-1 162,coding 320 amino acids.It was a complete full-length gene comparing with the homologues in GenBank.The protein had no transmembrane region.The protein contains no transmembrane region and subcellular location sequence.The predicted amino acid sequence showed 98% identity and 99% similarity with that of enolase from Schistosoma japonicum.Conclusion The cDNA sequence encoded enolase gene is screened from cDNA library of adult Taenia solium by bioinformatical method.The structure and character Taenia solium of the gene and protein of enolase gene are obtained.
