丙泊酚对内毒素血症大鼠肾脏功能及结构的影响 被引量:2

摘要 目的探讨丙泊酚对内毒素血症大鼠肾脏水通道蛋白-2(AQP2)、细胞间黏附分子-1(ICAM-1)、肿瘤坏死因子-α(TNF-α)基因mRNA和蛋白表达及对肾脏结构功能的影响。方法 Wistar大鼠48只随机分为6组,每组8只:对照1(C1)组、对照2(C2)组、内毒素(L)组、丙泊酚预处理(P1)组、丙泊酚同时给药(P2)组、丙泊酚后处理组(P3)组,模型制备成功12 h后,处死取材。观察肾组织病理学改变,检测肾脏功能学指标,RT-PCR、Western Blot及免疫组化方法测定AQP2I、CAM-1和TNF-α的表达。结果 L组较C1、C2组肾脏结构、功能学指标受损明显,AQP2 mRNA及蛋白表达减少I,CAM-1和TNF-αmRNA及蛋白表达增多;不同时间使用丙泊酚后,P1、P2、P3组肾脏结构、功能学指标受损程度较L组减轻,AQP2 mRNA及蛋白表达较L组增多I,CAM-1和TNF-αmRNA及蛋白表达较L组减少。结论丙泊酚可减轻内毒素血症肾组织的损伤,使AQP2表达增多,ICAM-1和TNF-α表达减少。
