莱芜猪GPx4基因在不同组织中的表达规律研究 被引量:3

摘要 为研究GPx4基因mRNA在莱芜猪不同组织中的差异表达规律,探讨该基因与地方猪种抗氧化性能的关系。本研究以5头莱芜猪为实验材料,提取10种组织的总RNA,应用实时荧光定量PCR技术,检测GPx4基因在莱芜猪不同组织的mRNA相对表达量。实时荧光定量PCR研究结果显示:GPx4基因在莱芜猪各组织中均有表达,组织之间的相对表达量存在显著差异(P<0.05);GPx4基因在莱芜猪脑、心、肝、脾等组织高丰度表达,并且极显著地高于其他组织中的表达量(P<0.01)。GPx4基因在不同组织中的表达水平由高到低依次为:肌肉>脊髓、大肠、背膘、小肠>肺,差异显著(P<0.05)。 The purpose of this investigation was to analyze GPx4 gene mRNA expression profiles of Laiwu pigs,and to evaluate the potential relationships between GPx4 mRNA relative expression and antioxidative capacity of Chinese native pigs.Five Laiwu pigs were used as experimental animal,and total RNA was isolated from 10 tissues of Laiwu pig.The mRNA expression levels of GPx4 gene of different tissues were measured using quantitative real-time PCR,and the results showed that it was tissue specific in GPx4 mRNA expression and the levels of mRNA expression were significantly difference between tissues.The GPx4 mRNA expression levels of brain,heart,liver and spleen were significantly higher than that of the other tissues(P0.01).The rank order of the GPx4 mRNA expression in the other tissues was muscle spinal cord,large intestine,backfat and small intestine lung,and there were significantly different(P0.05).
