摘要
从2 200多个海洋放线菌株中筛选得到菌株H41-26对花生青枯病菌(Pseudomonas solanacearum)有较强的拮抗作用。通过菌株形态特征、培养特性、生理生化特性、细胞壁化学组分及16S rDNA序列分析,初步确定菌株H41-26为链霉菌属金色(Aureus)类群中的新种-闸坡链霉菌(Streptomyces zhapoensis)。采用硅胶柱层析和高效液相半制备的分离纯化技术,主要活性成分鉴定为放线菌素V。首次报道了放线菌素V对花生青枯病菌的最小抑制浓度为3.91 mg/mL,8%放线菌素V乳油稀释1 000倍施药7 d和10 d后对花生青枯病的盆栽试验防治效果均在70%以上,具有较好的防治效果,为放线菌素V在植物病害防治方面的研究应用奠定了基础。
A strain H41-26 with strong inhibitory effect against pseudomonas solanacearum was screened from more than 2 200 strains of marine actinomycetes.Based on the study on its morphological,physiological and biochemical characteristics,Chemical compositions of cell wall,together with sequencing of 16S rDNA genes and phylogenetic analysis,the strain H41-26 was initially identified as Streptomyces zhapoensis.Using technologies of silica gel column chromatography and semi-preparative HPLC,the main active ingredient was identified as actinomycin V.It was first reported that minimum inhibitory concentration(MIC) of actinomycin V against P.solanacearum was 3.91 mg/mL.The pot experiment results show that actinomycin V had good control effect with more than 70% control effect of 8% actinomycin V EC diluted 1 000-fold after 7 days and 10 days treatments,so actinomycin V laid the foundation for its application in the plant diseases control research.
