异丙基肾上腺素充血性心衰大鼠模型NE、AngⅡ的改变 被引量:1

Changes of plasma NE and AngⅡin rats with isoproterenol-induced chronic heart failure

摘要 目的建立异丙基肾上腺素(ISO)诱导SD大鼠充血性心力衰竭(CHF)模型,观察血浆NE、AngⅡ的变化。方法应用大剂量ISO(170mg/kg)对SD大鼠皮下注射2次,建立CHF模型,造模后18w测量左室舒张末内径(LVEDD)、左室收缩末内径(LVESD)、室间隔厚度(IVST)、左室后壁厚度(LVPWT)、缩短分数(FS)、左室射血分数(LVEF);利用酶联免疫方法检测血浆NE和放免法检测血浆AngⅡ水平,并取心肌观察病理形态学特征。结果与对照组(CON)相比,ISO组大鼠心功能明显下降,血浆NE、AngⅡ显著升高;ISO组大鼠心肌细胞肥大,部分心肌细胞有嗜酸变性和(或)坏死,心肌间质结缔组织增生。结论应用大剂量ISO诱导大鼠CHF模型,该作用可能与血浆NE、AngⅡ升高有关。 Objective To establish a rat model of isoproterenol(ISO)-induced chronic heart failure(CHF),and to observe the changes of plasma NE and angiotensinⅡ(AngⅡ).Methods Male SD rats were randomly assigned into ISO-induced chronic heart failure group(ISO group,n=50)and healthy control group(CON group,n=10).Animals in ISO group received the subcutaneous injections of high dose of ISO(170 mg/kg)twice to establish the model of CHF and those in CON group were subcutaneously injected 0.25 ml of normal saline.18 w later,left ventricular end-diastolic dimension(LVEDD),left ventricular end-systolic dimension(LVESD),interventricular septal thickness(IVST),left ventricular posterior wall thickness(LVPWT),fractional shortening(FS)and left ventricular ejection fraction(LVEF)of both groups were measured by echocardiogram.The levels of plasma NE and AngⅡ were detected by ELISA.In addition,myocardial samples were obtained for the pathologic and morphologic observation.Results Compared with CON group,LVEF and FS decreased,LVEDD and LVESD enlarged and plasma NE and AngⅡincreased significantly in ISO group.Pathological examination showed that the cardiocytes of the animals in ISO group were hypertrophic,parts of them were acidophilic denaturation and/or necrosis,and the desmoplasia was found in myocardial interstitial tissues.Conclusion CHF model of rats can be induced by high dose of isoproterenol.The increased levels of plasma NE and AngⅡ may be involved in the induced roles.
