摘要
目的探讨丹参、维生素和牛磺酸对高同型半胱氨酸血症(HHcy)兔动脉粥样硬化的影响。方法采用高L-蛋氨酸饮食方法建立兔HHcy病理模型,用丹参、牛磺酸、叶酸、VB6、VB12作为干预药物,以正常家兔为对照,分别于实验0周、4周、8周检测各组血清同型半胱氨酸(Hcy)、丙二醛(MDA)、总胆固醇(TC)、甘油三酯(TG)、一氧化氮(NO)、血栓素B2(TXB2)含量和超氧化物岐化酶(SOD)活力,并通过血管超声和组织学观察腹主动脉血管病理变化。结果蛋氨酸组第4周、8周时血清Hcy、MDA、TG、TC、ET、TXB2含量显著高于同期NC组(P<0.01,P<0.05),而NO含量、SOD活力则明显降低(P<0.01);丹参和牛磺酸组Hcy显著高于对照组(P<0.01),维生素组显著低于蛋氨酸组与对照组无差异;与蛋氨酸组比较丹参、维生素和牛磺酸组MDA、TG、TC、ET、TXB2含量显著降低(P<0.01),而NO、SOD显著增高(P<0.01)。血管超声和组织学观察,蛋氨酸组腹主动脉内膜明显增厚、血管平滑肌细胞增生,弹力纤维断裂、排列紊乱、有斑块形成;丹参、牛磺酸和维生素组,血管内膜较光滑、结构排列整齐无斑块,与NC组比较差异无统计学意义。结论丹参和牛磺酸通过抑制Hcy诱导的氧化应激和细胞增值,调节血管舒缩及凝血与纤溶之间的平衡;维生素加速Hcy代谢,抑制高Hcy导致的血管损害,发挥抗AS形成作用。
Objective To investigate the effect of salvia miltiorrhiza, vitamin and taurine to atherosclerosis in high homocysteine(HHcy)rahhits. Methods HHcy rabbits models were established by high L-methionine diet. The salvia miltiorrhiza, taurine, folic acid, VB6 and VBlz were used as intervene drugs. The normal rabbits were as control group. The homoeysteine (Hcy), malonaldehyde(MDA), nitric oxide (NO) , thromboxane B2 (TXB2) and the activity of superoxide dismutase (SOD) were detected in each group on 0, 4, 8 weeks. The pathological changes of abdominal aortic were observed by vascular ultrasound and histology at the same time. Results Compared with control group, Hcy, MDA, TG, TC, ET and TXB2 levels in serum were significantly increased in the methionine group(P〈0.01 or P〈O. 05), but the level of NO and the activity of SOD were significantly decreased; compared with control group, Hey levels in the salvia miltiorrhiza group and taurine group were significantly increased; Hcy levels in the vitamin group were significantly lower than taurine group, but there was no significant difference compared with control group; compared with methionine group, the levels of MDA, TG, TC, ET, TXB2 were sig nificantly decreased in the salvia miltiorrhiza group, vitamin group and taurine group(P〈0.1), but the level of NO and the ac- tivity of SOD were significantly increased(P〈0.01). The vascular ultrasound and histology showed that, the abdominal aortic tunica intima thickened significantly, vascular smooth muscle cells hyperplasy, elastic fibers fractured, disordered and got plaque formed in the methionine group; the vascular intima were more smoothly, the structure aligned and no plaque in the sal- via miltiorrhiza group, taurine group and vitamin group, there was no significant difference compared with control group, Con- clusion Salvia miltiorrhiza and taurine regulate the vasomotor and the balance of coagulation and fibrinolysis by inhibiting the oxidative stress and cell proliferation caused by Hcy; vitamin can accelerate the metabolism of Hcy, inhibit the vascular impair caused by HHcy, and plays a role in the formation of anti AS.
