摘要
目的:通过建立H2O2诱导心肌细胞氧化应激损伤模型,观察GSTT对心肌细胞的保护作用。方法:本实验通过对大鼠乳鼠心肌细胞进行体外培养,采用H2O2制备心肌细胞氧化应激损伤模型,MTT法检测细胞存活率,荧光显微镜观察Hoechst33258染色凋亡心肌细胞形态,流式细胞术检测凋亡率及NF-κB含量。结果:蒺藜皂苷明显保护受损心肌细胞,降低细胞凋亡率;结论:蒺藜皂苷对心肌细胞具有保护作用。
Objective: Through the establishment of the model for the H202 induction myocardium cells oxidatie stress damage, observing the protective effect of myocardial cells GSTT. Method: Through the study, neonatal rat cardiomyocytes cultured in vitro, myocardium cells by H202 oxidative stress model, the survival ability was detected by Methyl thiazolyl tetrazolium (MTT) method. Cell apoptosis rate was detected by flowcytomety. The apoptosis cells morphous were observed by hoeehst33258 through fluorescence microscope. Result: GSTT significantly impaired myocardial protection and reduce the apoptosis rate. Conclusion: GSTT had protective effects on myocardium cells.
