摘要
建立紫外分光光度法测定豆腐柴叶中总黄酮含量的方法,研究测定波长、乙醇浓度、液料比、超声时间等因素对测定结果的影响,并在优化的试验条件下对不同生长期的野生豆腐柴叶中总黄酮含量进行测定,检出限为0.21μg/L,相对标准偏差小于3.5%,回收率为97.50%~102.50%。该法简便,快捷,灵敏度高,准确,适用于豆腐柴叶中总黄酮含量的测定。测定结果表明,野生豆腐柴叶中总黄酮含量与生长期有一定联系,为选择合适的豆腐柴叶采摘期提供了参考。
A spectrophotometry method has been established and validated for determination of flavonoids content of premna microphylla turcz.Operation conditions such as wavelength,concentration of alcohol,liquid ratio,ultrasonic time which have effects on determination results,have been investigated.The total flavonoids of wild premna at different growth were determined under the optimized experimental conditions,the detection limit was 0.21 μg/L,the relative standard deviation is less than 3.5% and the recovery was 94.25%~102.57%.The method is simple,fast,high sensitivity and accurate,and was suitable for the quantitative analysis of flavonoids in premna microphylla turcz.The determination results showed that there are certain links between the flavonoids content of wild premna and the growth period,it also provides a reference for the selection of a suitable picking period of Premna.
