摘要
本文采用聚丙烯酰胺凝胶电泳技术研究了吡虫啉对不同发育阶段星豹蛛酯酶同工酶活性的影响。结果表明,星豹蛛酯酶同工酶酶带可分为三个区域,其中EST-3区的酶带颜色最深,酶活性最强;EST-1区和EST-2区酶带颜色较浅,酶活性较弱。星豹蛛不同发育阶段酶带数由多到少的顺序为:雌成蛛>卵袋>雄成蛛>若蛛,酶活性由强到弱的顺序为:雌成蛛>雄成蛛>若蛛>卵袋。农药处理星豹蛛后,其酯酶同工酶活性受到了不同程度的抑制,且随着吡虫啉浓度的增大,同一发育阶段和同一性别的星豹蛛酶带的颜色逐渐变浅,边缘逐渐变模糊。在体和离体施药方法对酶活性的影响研究结果可知,在体施药与对照组差异不显著;离体施药后,只在EST-3区出现酶带而且颜色较浅,说明离体施药对星豹蛛酯酶同工酶酶活性的抑制作用较在体更强。研究结果表明,蜘蛛施用杀虫剂后通过抑制酯酶同工酶对毒物的消化,从而降低杀虫剂对其毒害作用,提高耐药能力。本研究结果为探讨蜘蛛对杀虫剂的抗性生化机制,合理使用杀虫剂以及协调生防与化防的关系具有重要意义。
A comparative studywas conducted to the esterase(EST) isozyme inPardosa astrigera at different develop- ment stages under application of imidacloprid pesticide by usirig the vertical slab polyacrylamide gel electrophoresis tech- nology. The results showed that the EST isozyme bands were divided into three areas. The color of bands in the EST-3 area was darker and the activity stronger than those in the EST-1 area and EST-2 area. The orders of the EST isozyme band number and activity at different development stages are respectively : female adult 〉 egg bag 〉 male adult 〉 larval and female adult 〉 male adult 〉 larval 〉 egg bag. The activity of EST isozyme was inhibited after being treated with imidacloprid to some degrees. With the increase of imidacloprid concentration, the number of bands decreased, color shoaled, edge fogged. With the results of study on the effects of EST isozyme activity by the application of different methods of pesticide, in vivo, there was no significant difference from control group ; in vitro, merely shoaled bands inthe EST-3 area after being compared with the control group, which showed that EST isozyme activity was resisted more in vitro than in vivo, and Pardosa astrigera reduced the effect of poison with pesticide and increased their resistance through the restraint of the digesting ability of EST isozyme. The results have great significance for the discussion on biochemical mechanism of spider resistance to pesticide, for the reasonable use of pesticide and for the balance of relationship be- tween biocontrol and chemical control.
