神经肽Y系统在高剂量咖啡因、可可碱诱导的大鼠焦虑行为中的作用机制 被引量:5

摘要 目的:探讨咖啡因、可可碱对大鼠焦虑行为的影响及神经肽Y系统在其中的作用机制。方法:30只成年雄性Wistar大鼠,随机分为5组。分别予以生理盐水、咖啡因30mg/kg、咖啡因60mg/kg、可可碱30mg/kg、可可碱60mg/kg灌胃给药。给药后30min测试大鼠焦虑行为及检验海马神经肽Y-mRNA、Y1-mRNA、Y2-mRNA水平。结果:(1)较之对照组,咖啡因60mg/kg组TO、EO、EC、TE(F=21.774,P=0.001)、EO%(χ2=5.336,P=0.044)、TO%(χ2=7.750,P=0.019)均明显减低,差异有统计学意义;而可可碱30mg/kg组在各个指标上均明显增高(P<0.05)。(2)咖啡因60mg/kg组NPY-mRNA的表达增高(F=13.961,P=0.004),而可可碱30mg/kg组NPY-mRNA明显降低(F=7.597,P=0.020),Y1-mRNA(F=10.541,P=0.009)、Y2-mRNA(F=4.532,P=0.040)的表达明显增高,差异有统计学意义。结论:超高剂量的咖啡因具有致焦虑作用,而高剂量的可可碱有抗焦虑作用,神经肽Y系统与他们的作用机制有关。
