高效液相色谱法同时测定多维生素糖浆中5种维生素和山梨酸含量

摘要 目的建立可同时测定多维生素糖浆中抗坏血酸(VitC)、烟酰胺(VitPP)、盐酸硫胺(VitB1)、核黄素-5'-磷酸钠、盐酸吡多辛(VitB6)及防腐剂山梨酸含量的高效液相色谱法。方法采用高效液相色谱-紫外分光光度法,使用Allthna C18柱(250 mm×4.6 mm,5μm),流动相A为0.01 mol/L己烷磺酸钠溶液(1 000 mL溶液加入三乙胺0.25 mL和醋酸9.5 mL),流动相B为甲醇(梯度洗脱),流速为1.0 mL/min,检测波长为280 nm。结果 VitC,VitPP,VitB1、核黄素-5'-磷酸钠、VitB6、山梨酸的质量浓度分别在0.15~2.0 g/L,0.1~1.0 g/L,0.02~0.4 g/L,0.02~0.4 g/L,0.007~0.1 g/L,0.03~0.6 g/L范围内与峰面积线性关系良好;平均回收率分别为99.9%,98.4%,99.4%,100.9%,100.4%,100.3%,RSD=0.5%,0.6%,0.4%,0.9%,1.0%,0.8%(n=9)。结论该方法快速、灵敏,色谱峰分离度良好,结果准确可靠,可作为多维生素糖浆的质量控制项目之一。 Objective To develop a HPLC method for the simultaneous determination of vitamin C(VitC),vitamin PP(VitPP),vitamin B1(Vit B1),riboflavin-5′-sodium phosphate,vitamin B6(VitB6) and sorbic acid in Alvityl Syrup.Methods After being diluted with pure water,the samples were separated by HPLC on the Allthna C18 column(250 mm×4.6 mm,5 μm)with the mobile phase A of 0.01 mol/L sodium hexanesulfonate aqueous solution(1 000 mL solution containing 0.25 mL triethylamine and 9.5 mL acetic acid) and the mobile phase B of methanol.The mobile phases were gradiently eluted.The flow velocity was 1.0 mL/min.The detection wavelength was 280 nm.Results The calibration curves for VitC,VitPP,VitB1,riboflavin-5′-sodium phosphate,VitB6 and sorbic acid were linear over the range of 0.15-2.0 g/L,0.1-1.0 g/L,0.02-0.4 g/L,0.02-0.4 g/L,0.007-0.1 g/L and 0.03-0.6 g/L,respectively And the average recovery rates were 99.9%,98.4%,99.4%,100.9%,100.4%and 100.3%,RSD=0.5%,0.6%,0.4%,0.9%,1.0%,0.8%(n=9)respectively.Conclusion The assay is proved to be sensitive,accurate and convenient with good resolution,which can be applied to control the quality of Alvityl Syrup.
