摘要
目的通过研究消幻汤对精神分裂症模型自主活动、空间学习记忆及纹状体内五羟色胺水平的影响,探讨消幻汤治疗精神分裂症的可能机制。方法雌性清洁级sD大鼠,随机分为:正常组,模型组,消幻汤组,利培酮组。按照0.1mg/kg给予地卓西平马来酸盐(MK-801)连续腹腔注射两周,刻板行为、旷场实验及Morris水迷宫实验评价大鼠行为的改变。消幻汤组给予消幻汤12g/4ml/kg,利培酮组给予利培酮0.2mg/kg之后,余两组灌服等量生理盐水。4周后观察大鼠行为改变,并使用库仑阵列电化学高效液相色谱测定纹状体内神经递质含量的变化。结果模型组的大鼠刻板行为评分[(2.54±0.52)分,(2.42±0.51)分,(2.30±0.48)分],高于正常组评分[(1.08±0.28)分],符合模型制备成功标准。经药物干预后,消幻汤组、利培酮组大鼠自主活动[(780.57±248.99)分,(819.76±267.21)分]低于模型组[(1204.15±344.25)分],差异有显著性(P〈0.05);消幻汤组逃避潜伏期[(30.25±13.67)S]较模型组[(44.72±19.88)S]缩短,穿台次数[(5.67±1.44)次]多于模型组[(3.08±1.44)次],均差异有显著性(P〈0.05)。消幻汤组及利培酮组纹状体内五羟色胺(5TH)含量[(0.08±0.26)μg/g,(0.11±0.07)μg/g]较模型组[(0.25±0.14)μg/g]降低(P〈0.05),消幻汤组五羟吲哚乙酸(5HIAA)含量[(0.28±0.36)μg/g]较模型组[(0.27±0.09)μg/g]差异无显著性(P〉0.05)。结论消幻汤改善了精神分裂症模型大鼠刻板行为、自主活动和学习记忆损伤,其机制可能与参与了纹状体内的五羟色胺含量的调节有关。
Objective To study the effects of Xiaohuan Decoction on the locomotor activity,spatial learning and memory,and the serotonin(5-HT) in striatum in schizophrenia model rat, and the possible mechanism of Xiaohuan Decoction on schizophrenia. Methods The rats were randomly assigned into four groups: normal group, model group, Xianhuan Decoction group and risperidone group; The schizophrenia model was established by intraperitoneal injection 0. 1mg/kg MK801 for 2 weeks except the normal group, and then evaluated by open field test and water maze. Xianhuan Decoction group were given 12 g/kg Xianhuan Decoction and risperidone group were given 0.2 mg/kg risperidone for 4 weeks respectively,the other groups were treated with placebo. The open field test and electrochemical coulometric array HPLC were used to detect the relevant data after 4 weeks. Results The score of stereotyped behavior in model rats was ( 2.54 ± 0.52,2.42 ± 0.51,2.30 ± 0.48 ) , it was higher than the normal group( 1.08 ± 0.28) ,the models were successful according the standard. After drug intervention,the locomotor activity of Xiaohuan Decoction and risperidone group (780.57 ±248.99,819.76±267.21 ) was less than the model group (1204.15 ± 344.25 ) , there was a significant difference (P 〈 0.05 ) ;and escape latency latency of Xiaohuan Decoction (30.25 ± 13.67) was shorter than that the model group (44.72 ± 19.88 ) , the number of units to wear (5.67 ±1.44) was more than thai the model group (3.08 ±1.44) ,there were significant difference (P 〈 0.05 ). The content of 5-HT in striatum were significantly decreased in Xianhuan Decoction group (0.08 ± 0.26) and risperidone group (0.11 ± 0.07 ) compared with model group ( 0.25 ± 0.14 ) (P 〈 0.05 ) , and there was no significant difference of the content of 5-HIAA between Xianhuan Decoction group(0.28 ± 0.36) and model group(0.27 ± 0.09) (P 〉 0.05 ). Conclusion Locomotor activity and memory impairment were improved by Xianhuan Decoction, and the possible mechanism may be that Xianhuan Decoction can regulate the content of 5-HT in stfiatum in the schizophrenia model rat treated.
