摘要
目的:观察万米跑后不同时相人体外周血淋巴细胞凋亡及血液中超氧化物歧化酶(SOD)、谷胱甘肽(GSH)和丙二醛(MDA)活性的变化,探讨大强度耐力运动后血液淋巴细胞凋亡的机制。方法:选取8名参加运动会万米长跑的体育专业男运动员,分别在运动前1天(RG)、运动后2h、24h和48h采血抗凝,分离淋巴细胞,用流式细胞仪检测血液淋巴细胞凋亡率,并测定血清中SOD活性、GSH和MDA含量。结果:运动后2h和24h外周血淋巴细胞凋亡率显著高于安静水平,2h显著高于24h;48h基本恢复安静水平。2h血浆中SOD酶活性、GSH和MDA含量均较安静水平显著升高;24h SOD和MDA仍显著高于安静水平,而GSH水平则无显著差异;48h后SOD、GSH和MDA均较安静水平无显著差异。外周血MDA含量与淋巴细胞凋亡率呈正相关(r:0.757;P:0.001)。结论:万米长跑对外周血淋巴细胞凋亡率有诱发效应,而凋亡率随时间的变化呈现"开窗理论"特征。运动引起的机体自由基代谢失衡可能是淋巴细胞凋亡率变化的原因之一。
object: we investigated the lymphocytes apoptosis and the level of anti-oxidative status on human serum in differ- ent times after ten-thousand-meter running, and discussed the mechanism of lymphocytes apoptosis induced by endurance exercise. Method:We selected eight healthy male athletes, who participated in ten-thousand meter running in sports games. The subjects were tested twenty-four hours before exercise and 2 hours,24hours and 48 hours after exercise. We investiga- ted the lymphocytes apoptosis on human peripheral blood by flow cytometry and detected the level of SOD, GSH and MDA in serum. Results:The results have showed that the lymphocytes apoptosis on peripheral blood cell significantly increased 2h and 24h after exercise and 2h it was significant higher than 24h. It reduced to the level of pre-exercise after 48 hours. The activity of SOD and the content of GSH and MDA in serum increased significantly at 2h compared with the pre-exer- cise, and SOD activity and MDA content were still significant higher than the pre-exercise. The changes of the indexes in serum at 48h had no significant difference compared with pre-exercise. A negative correlation was found between MDA and the rate of lymphocyte apoptosis. Conclusions : ten-thousand-meter running induces have effects on rates of peripheral blood lymphocytes, and the apoptosis rate changes over time show "open windows" feature. Exercise -induced metabolic imbal- ance of free radicals might be one of the lymphocytes apoptosis mechanisms.
