摘要
目的探讨促红细胞生成素(EPO)对大鼠坐骨神经损伤后背根神经节细胞Bcl-2和Bax表达的影响及其作用机制,为周围神经损伤的临床治疗提供实验依据。方法♀SD大鼠36只,制备大鼠左侧坐骨神经缺损模型,随机分为3组:EPO组、神经生长因子(NGF)组和生理盐水(NS)组。3组分别腹腔注射EPO、NGF和NS。术后第7、14天对每组大鼠进行一般观察,并采用免疫组化法检测L5背根神经节中Bcl-2和Bax的表达。结果术后第7、14天,EPO组Bcl-2的表达高于NS组(P<0.05,P<0.01),与NGF组比较,差异无显著性。EPO组Bax的表达低于NS组(P<0.01),与NGF组比较,差异无显著性。结论 EPO可能通过促进Bcl-2的表达,抑制Bax的表达,对大鼠坐骨神经损伤发挥保护作用。
Objective To explore the effect of erythropoietin(EPO) on Bcl-2 and Bax expression of dorsal root ganglion after sciatic nerve injury in rats and provide experimental basis for clinical treatment of peripheral nerve injury.Methods 36 female SD rats were set up the model of the sciatic nerve defect,then the rats were divided into three groups: NS group,EPO group and nerve growth factor(NGF) group,which were injected intraperitoneally with normal saline,erythropoietin and NFG,respectively.At 7 and 14 days after operation,the rats were taken for morphological assessment.The expression of Bcl-2 and Bax of L5 dorsal root ganglion were observed by immunohistology.Results At 7 and 14 days after operation,the expression of Bcl-2 in EPO group was more than that in NS group(P0.05,P0.01),while the expression of Bax in EPO group was less than that in NS group(P0.01).There was no significant difference between the EPO group and NGF group.Conclusion EPO can promote the expression of Bcl-2 and inhibit the expression of Bax,which may provide protective effect on sciatic nerve injury in rats.
