摘要 探讨小鼠纹状体内注射6-OHDA制作PD模型可行性。方法将6-OHDA注射入小鼠单侧纹状体内,使用阿朴吗啡和安非他命诱发的动物单侧旋转进行行为学检测,同位素法检测皮层、左右纹状体内多巴胺能神经元标志酶酪氨酸羟化酶(tyrosine hydroxylase,TH)的活性。结果与结论小鼠纹状体内注射6-OHDA后,可成功制作急性PD模型,成功率为95%,模型持续时间大约4周。 Aim Until now, there is no data that show mouse microinjeetion 6-hydroxydopamine (6-OHDA) for Parkinson' s disease (PD) model in our country. Here a mouse PD model was suggested by injection 6-OHDA into unilateral striatum. Methods The right striatum of mice was injected 6-OHDA, the Apomorphine- and Amphetamine-induced rotation behaviors were measured, and the tyrosine hydroxylase (TH) activity of cortex,left and right striatum were assayed by radioassay method. Re- suits and Conclusion The results indicate that mouse injected 6-OHDA into striatum can be used to induce acute PD animal model, the successful rate was 95%, and the period is about four weeks.
