内毒素血症大鼠肾脏水通道蛋白2等基因表达及肾脏结构功能的改变 被引量:6

The Expression AQP2 Protein and Alteration in Renal Morphology and Function in Kidney of Endotoxemic Rats

摘要 目的:探讨内毒素血症大鼠肾脏水通道蛋白(AQP)2、细胞间黏附分子(ICAM)-1、肿瘤坏死因子(TNF)-α mRNA和蛋白表达及肾脏结构功能的改变。方法:Wistar大鼠24只随机分为空白对照1(C1)组,只注射生理盐水和脂肪乳的对照2(C2)组及注射内毒素和脂肪乳的内毒素(L)组,每组8只,在内毒素血症模型造模成功12h后,处死取材。观察肾组织病理学改变,RT-PCR、Western blot及免疫组化方法测定AQP2、ICAM-1和TNF-α的表达。结果:L组较C1组、C2组肾组织出现病理损伤,尿量减少,尿渗透压下降,血渗透压、血肌酐及尿素氮增高,AQP2 mRNA及蛋白表达减少,ICAM-1、TNF-α mRNA及蛋白表达增多,差异均有统计学意义(均P<0.001)。结论:大鼠内毒素血症可导致肾组织形态改变和功能损伤,AQP2表达减少,ICAM-1和TNF-α表达增多。 Objective:To investigate the expression of aquaporin-2(AQP2),intercellular adhesion molecule(ICAM)-1 and tumor necrosis factor(TNF)-α mRNA and alterations in renal morphology and function of endotoxemic rats.Methods:Twenty-four Wistar rats were randomly divided into 3 groups(n=8 for each group),C1 group(sham control group),C2 group(saline and fat milk group),and L group(endotoxin and fat milk group).The morphology and function of kidney were observed and levels of AQP2,ICAM-1,and TNF-α were detected with RT-PCR and Western blot 12 h after rat model establishment.Results:There were significantly changes in morphological damage and functional impairment,and downregulation of AQP2 mRNA and protein expression and upregulation of ICAM-1 and TNF-α mRNA and protein expression in rats of L group than those of C1 group and C2 group(P 0.001).Conclusion:Endotoxemia led to the renal damage,decreased levels of AQP2 and increased levels of ICAM-1 and TNF-α in rat models.
