放疗后椎体骨细胞放射性损伤的评估 被引量:3

摘要 目的探讨检测放疗后椎体骨细胞损伤的有效方法。方法 20只比格犬随机分为两组:放疗组16只,在10~14d内,犬胸T9~10椎体给予生物剂量40~70Gy的放疗;空白对照组4只。放射治疗结束3个月后,取T9~10椎体骨。骨组织分成2份,分别应用单细胞凝胶电泳技术(彗星实验)和免疫组化方法检测椎体骨细胞损伤情况。结果在彗星实验结果显示,放疗损伤程度与放射剂量之间有明显的剂量-效应关系,而免疫组化的结果无法推算出上述剂量-效应关系。结论彗星实验检测放疗后骨损伤,具有简便、灵敏、准确、重现性好的特点,可应用于放疗后椎体骨细胞放射损伤的检测。 Objective To investigate the effective method for evaluating radiation damages of vertebral bone cells after radiotherapy.Methods Twenty dogs were randomly divided into 2 groups of A(16 dogs) and B(4 dogs,blank control).Dogs in group A were treated with mimic radiotherapy of 40-70 Gy at T9-10 vertebral body for 10 to 14 days.The sample of T9-10 vertebral body from each dog was taken out and divided to two pieces for determining cell DNA damages with single cell gel electrophoresis assay and immunohistochemisty,respectively.Results The dose-effect relationship was shown by single cell gel electrophoresis assay,which was not by immunohistochemisty.ConclusionThe single cell gel electrophoresis assay can be used to evaluate the radiation damages of vertebral bone cells after radiotherapy with the advantages of simplicity,sensitivity,accuracy and repeatability.
