人肥大细胞羧肽酶真核表达重组质粒DNA的免疫原性观察

摘要 目的观察人肥大细胞羧肽酶(hMC-CP)真核表达重组质粒DNA的免疫原性。方法以分泌性和非分泌性hMC-CP真核表达重组质粒(pcDNA3.1/S-CP,pcDNA3.1/CP)DNA为免疫原,分别以皮下及肌肉两种途径免疫BALB/c小鼠,间隔免疫时间2周,共免疫3次;末次免疫后第7天,采用ELISA法检测血清中特异性抗体的产生。结果以pcDNA3.1/S-CP、pcDNA3.1/CP DNA免疫小鼠后,都能诱导产生抗hMC-CP抗体,而pcDNA3.1空载体和NS则不能。通过肌注途径,以pcDNA3.1/S-CP免疫小鼠所得血清抗体效价均值为1∶6 400,以pcDNA3.1/CP免疫小鼠所得血清抗体效价均值为1∶3 200,两者比较,P>0.05;通过皮下注射途径,以pcDNA3.1/S-CP、pcDNA3.1/CP DNA免疫小鼠所得的免疫血清抗体效价均值均为1∶3 200,两者比较,P>0.05;而同一载体肌肉与皮下免疫,所得血清抗体效价比较,P>0.05。结论 hMC-CP真核表达重组质粒DNA具有较好的免疫原性,表达质粒是否为分泌性表达不影响其免疫原性,其免疫原性也不受质粒DNA注射途径的影响。
