摘要
目的:通过观察人类重组白介素-1受体拮抗剂(IL-1ra)干预应力屏蔽后大鼠跟腱形态学变化,了解IL-1在肌腱挛缩中的影响及IL-1ra对肌腱挛缩的干预作用。方法:40只SD大鼠随机分为A、B、C和D组各10只,另从C、D组中分别抽取5只大鼠以右后肢为正常对照组(E组)。将前4组大鼠均实行左跟腱完全应力屏蔽造模术;术后即刻B、D组使用5μg IL-1ra、A及C组使用0.1ml磷酸缓冲盐溶液(PBS)于大鼠左后跟腱周围注射。于1、2及3周时A及B组分别再注射1次,C及D组于术后1周时再注射1次。干预2及4周时肉眼下及透射电镜下观察跟腱形态学改变。结果:肉眼下B和D组跟腱明显比A和C组细小、光滑,但较E组粗。电镜下2周时A和C组胶原原纤维较B和D组疏松、紊乱;4周时A和C组横切面和纵切面均与E组差异有统计学意义,而B和D组横切面与E组接近,但纵切面略显松弛。结论:IL-1能明显预防应力屏蔽所致的肌腱挛缩。
Objective:To investigate the effects of interleukin-1(IL-1) on tendon contracture and the preventive effects of interleukin-1 receptor antagonist(IL-1ra) on tendon contracture by observing the morphological changes of the stress-shielded achilles tendons after the intervention with IL-1ra.Methods:Forty SD rats were randomly divided into the 2-week PBS group,the 2-week IL-1ra group,the 4-week PBS group,the 4-week IL-1ra group and the normalcontrol group.IL-1ra was injected around the achilles tendons in the IL-1ra groups.The achilles tendons were tested morphologically under a transmission electron microscope(TEM).Results:On gross examination,the achilles tendons in the IL-1ra groups were significantly smoother and smaller than those of the PBS groups,but thicker than those of the normal control group.Under a TEM,the collagen fibrils of the 2-week PBS group were looser and more disorder than those of the 2-week IL-1ra group.The collagen fibrils of the 4-week PBS group were significantly different from those of the normal control group in longitudinal section and cross section.The collagen fibrils of the 4-week IL-1ra group were similar to those of the normal control group in cross section,but slightly looser than those of the normal control group in longitudinal section.Conclusion:IL-1 plays an important role in tendon contracture caused by stress shielding.IL-1ra can prevent tendon contracture.
