胞外不同钠浓度对培养新生小鼠心肌细胞钠电流的影响

摘要 目的:探讨细胞外液不同的钠浓度对培养新生小鼠心肌细胞Na+电流的影响。方法:采用膜片钳技术,在浴液不同的钠离子浓度下,记录培养新生小鼠的心肌细胞钠电流的激活、失活以及恢复曲线。结果:相对于低钠浴液,高钠浴液记录的培养新生小鼠心肌细胞钠电流明显延迟,电流幅度增加也不明显,激活曲线不圆滑。结论:胞外高钠不是记录钠电流最合适的实验方案。 Objective: To study the effect of different extracellular sodium concentration on Na+ current in cultured cardiomyocytes of suckling mouse.Methods: Using the patch clamp technique of the whole cell configuration,with the different concentration of sodium in bath solution,Na+ current of activation,inactivation and recovery curve were recorded.Results:Compared with low sodium bath solution,Na+ current was significantly delayed,the increase of current was not obvious,and Ⅰ-Ⅴ curve was not smooth in high sodium bath solution.Conclusion: Extracellular solution with high sodium is not appropriate to record Na+ current..
