摘要
目的研究氯化两面针碱与端粒G-四链体的相互作用。方法采用圆二色(CD)光谱和荧光滴定法测试氯化两面针碱与人端粒DNA序列d[G3(T2AG3)3]的相互作用。结果与结论CD光谱表明氯化两面针碱在体外能够诱导人端粒DNA序列d[G3(T2AG3)3]形成反平行G-四链体,并能提高G-四链体的热稳定性;荧光滴定实验表明化合物与G-四链体具有很好的亲和力。
Objective To investigate the interaction between nitidine chloride and G-quadruplex.Methods Circular dichroism(CD) spectrum and fluorescence titration were employed to analyze the interaction between human telomeric DNA d[G3(T2AG3)3] and nitidine chloride.Results and Conclusion CD spectrum showed that nitidine chloride could induce human telomeric DNA d[G3(T2AG3)3] to form and stabilize anti-parallel G-quadruplex in vitro and good affinity between compound and DNA was demonstrated by fluorescence titration.
