MCI-186对脊髓损伤大鼠炎症因子释放及神经营养因子表达的影响

摘要 目的:研究MCI-186对脊髓损伤(spinal cord injury,SCI)大鼠炎症因子的释放及神经营养因子表达的影响。方法:建立大鼠脊髓半切SCI模型,随机分成两组,分别予MCI-186或磷酸盐缓冲液(PBS)治疗,在术后1、3、7 d和14 d动态测定受损伤脊髓组织丙二醛(MDA)超氧化物歧化酶(SOD)的含量以及脑原性神经生长因子(BDNF)mRNA的表达,ELISA法测定血清及受损伤脊髓局部组织白介素-6(IL-6)、白介素-8(IL-8);神经营养因子-3(NT-3)、BDNF蛋白表达。并同时与假手术组对比。结果:与模型组比较,MCI-186能显著降低SCI大鼠血清炎性因子IL-6、IL-8的浓度,促进脊髓组织NT-3、BDNF表达。结论:MCI-186可抑制炎症因子释放,增加内源性神经营养因子表达。 Objective: To investigate the the protective effects of MCI-186 on the release of inflammatory cytokines and expression of neurotrophic factors in spinal cord injuried (SCI) rats. Methods: SCI model was reproduced by hemisection, Divided into two groups, the model immediately after a successful start to treat with the MCI-186 or phosphate buffered saline (PBS), after 1,3,7 d and 14 d dynamic determination of injured spinal cord tissue malondialdehyde (MDA) and superoxide dismutase (SOD) levels and brain derived neurotrophic factor (BDNF) mRNA expression, ELISA method was used to measure serum and spinal cord injury in the local tissue by interleukin-6(IL-6), interleukin-8(IL-8). NT-3, BDNF protein. And also compared with the sham group. Results: Compared with model group, MCI-186 could significantly reduce serum levels of inflammatory cytokines IL-6, IL-8 and upregulate the expression of NT-3, BDNF in the spinal cord after injury. Conclusion: MC1-186 can inhibit the inflammatory cytokine release, increased endogenous neurotrophic factor expression
