摘要
目的探讨Syk基因在子宫内膜良、恶性组织中的表达及其启动子甲基化与子宫内膜癌发生、发展的关系。方法采用RT-PCR方法检测52例子宫内膜癌及癌周组织、30例子宫内膜不典型增生、28例复杂型増生、30例单纯型增生和40例正常内膜组织中Syk mRNA的表达情况,同时采用MSP方法检测其基因启动子甲基化情况。结果在子宫内膜癌组、癌周组织、不典型增生组、复杂和单纯型增生及正常子宫内膜组中Syk基因表达依次增高,Syk基因甲基化程度依次降低,差异有统计学意义(P<0.05);Syk基因表达水平及其基因启动子甲基化程度在复杂型增生、单纯型增生及正常子宫内膜间的差异无显著性(P>0.05);子宫内膜癌中Syk基因的阳性表达及甲基化程度与癌症的临床分期(χ2=4.65,P<0.01;χ2=4.33,P<0.05)、淋巴结转移(χ2=10.68,P<0.01;χ2=5.86,P<0.05)、肌层浸润深度(χ2=5.82,P<0.05;χ2=3.90,P<0.05)相关,而与内膜癌组织学分级无明显相关性(χ2=0.03,P>0.05;χ2=0.94,P>0.05)。Syk基因启动子甲基化与Syk基因表达缺失明显相关(P<0.05)。结论 Syk基因启动子甲基化可能是导致Syk基因失活的原因之一,Syk基因启动子甲基化可能与子宫内膜癌发生发展有关。
Objective To explore the expression of Syk gene in benign and malignant tissue of endometrium,the relationship between promoter methylation of those tissue and the development of endometrial cancer is also investigated.Methods Syk mRNA was detected by RT-PCR in 52 cases of endometrial cancer and adjacent tissues,30 cases of endometrial atypical hyperplasia,28 cases of complex hyperplasia,30 cases of simple hyperplasia and 40 cases of normal endometrial tissue.Methylation of Syk promoter in all cases were detected by MSP methods.Results Follwing the groups of endometrial cancer,adjacent cancer tissue,endometrial atypical hyperplasia,complex hyperplasia,simple hyperplasia and normal endometrial tissue,expression of Syk gene was increased and promoter methylation was decreased.There was a significant difference(P0.05).There were no significant difference in the groups of complex and simple hyperplasy,normal endometrial tissue,in respect of expression and promoter methylation of Sky gene.In endometrial cancer groups,expression and promoter methylation of Syk gene were positively correlated with clinical stage of cancer(χ2=4.65,P0.01;χ2=4.33,P0.05),lymph node metastasis(χ2=10.68,P0.01;χ2=5.86,P0.05),depth of myometrial invasion(χ2=5.82,P0.05;χ2=3.90,P0.05),but wasn't correlated with histological grade of endometrial cancer(χ2=0.03,P0.05;χ2=0.94,P0.05).Promoter methylation of Syk gene was positively correlated loss expression of Sky gene.Conclusion Promoter methylation of Syk gene may be one reason for inactivation of it,which may cause the occurrence and development of endometrial cancer.
