摘要
目的检测维生素K2对肝癌细胞株内的肝癌衍生生长因子(HDGF)表达的作用。方法半定量逆转录.聚合酶链反应(RT-PCR)法检测0、10、30μm维生素l(2干预后肝癌细胞中HDGF的mRNA表达,Western blot法检测不同浓度维生素处理后的肝癌细胞中HDGF蛋白表达。结果RT.PCR结果示96h后10、30μm维生素K2干预组能呈剂量依赖性抑制肝癌细胞中HDGF的mRNA表达。Western blot法检测结果示96h后的10、30μm维生素K2干预组中HDGF的蛋白表达分别为无药对照组的(0.915±0.025)倍和(0.530±0.030)倍,差异有统计学意义(P〈0.01)。结论维生素K2可下调肝癌细胞中的HDGF表达。
Objective To investigate the regulation of hepatoma-derived growth factor (HDGF) expression by vitamin K2. Methods Semi-quantitative reverse-transcription-polymerase chain reaction (RT-PCR) was used to detect the mRNA levels of HDGF after exposure to 0, lO and 30μmoL/L vitamin K2 for 96 h, and Western blotting to detect the protein level of HDGF. Results RT-PCR revealed that vitamin K2 could inhibit the HDGF mRNA in a dose-dependent manner. The HDGF protein expression was siguificandy suppressed in 10 and 30 μmol/L vitamin K2 groups [ (0. 915 ±0. 025) and (0. 530 ±0. 030) respectively ] as compared with the control group (P 〈 0. 01 ). Conclusion Vitamin K2 inhibits the proliferation of hepatocellular carcinoma ceils through down-regulating HDGF expression.
