慢性乙肝脾肾阳虚证血浆蛋白质组学分析 被引量:6

摘要 目的:通过对比分析慢性乙肝脾肾阳虚证与正常健康对照组的血浆蛋白质组表达谱,初步探讨参与慢性乙肝脾肾阳虚证病理生理过程的分子机制。方法:采用双向电泳技术结合质谱分析等生物信息学分析方法对慢性乙肝脾肾阳虚证及正常健康对照组的血浆全蛋白进行比较蛋白质组学分析。结果:获得了8个有明显规律性变化的差异表达蛋白点,通过质谱分析鉴定获得7个蛋白质(蛋白点5、6为同一蛋白质):载脂蛋白C2(APO-C2)、玻连蛋白(vitronectin,VN)、结合珠蛋白(haptoglobin,HPT)、甲状腺素结合蛋白(transthyretin,TTHY)、载脂蛋白A2(APO-A2)、血清淀粉蛋白P(serum amyloid P-component,SAMP)、视黄醇结合蛋白(retinol binding protcin,RBP),其中APO-C2蛋白点表达量有所升高,其余6个差异蛋白点表现为明显下调趋势。结论:以上差异表达蛋白质的组合性变化可能是慢性乙肝脾肾阳虚证的物质基础,具有潜在的作为慢性乙肝脾肾阳虚证诊断、预后标记物或治疗靶点的意义。 Objective: To explore the molecular mechanism of Yang deficiency of spleen and kidney in chronic HBV by comparing plasma protein expression between patients with Yang deficiency of spleen and kidney in chronic HBV and normal controls.Methods: Plasma proteins from all experimental subjects were separated by two-dimensional gel electrophoresis(2-DE).The proteomic expression profiles were analyzed by comparative proteomics technique to discover important protein spots with different expressions and identify them by MALDI-TOF-MS.Results:Seven differential proteins were obtained: APO-C2,vitronectin,haptoglobin,transthyretin,APO-A2,serum amyloid P-component,and retinol binding protcin.Of the seven,APO-C2 showed higher expression level,while the other six proteins showed lower expression level in patients with Yang deficiency of spleen and kidney in chronic HBV.Conclusion: All proteins identified by this approach may be related to the material foundation of Yang deficiency of spleen and kidney in chronic HBV,and may be useful as molecular targets for Yang deficiency of spleen and kidney in chronic HBV diagnostics and therapeutics.
