摘要
目的:选择13种巴基斯坦传统草药,观察其对PC12细胞增殖的影响,并探讨其对H2O2、谷氨酸诱导PC12细胞损伤的保护作用,为寻找神经细胞保护作用新药提供实验依据。方法:体外培养PC12细胞,应用不同浓度的H2O2、GLU,分别建立神经细胞损伤模型,采用MTT法检测细胞存活率。结果:虎爪豆、诃子等能明显促进PC12细胞增殖作用(P<0.05);在H2O2 150μmol.L-1损伤模型中,药物组和模型组相比,罗勒、仙人掌、芦笋、灯油藤、诃子等能显著减轻H2O2的细胞损伤作用(P<0.05);在GLU 30mmol.L-1诱导PC12细胞损伤,与模型组比较,凤仙花、仙人掌、虎爪豆、诃子等均显示出损伤保护作用(P<0.05)。其中,诃子不仅能促进PC12细胞增殖,且在两种PC12细胞损伤模型中均具有较强的损伤保护作用。结论:具有促进PC12细胞增殖的药物为诃子、罗勒、虎爪豆;具有对抗H2O2损伤作用的药物有:诃子、凤仙花、仙人掌、芦笋、灯油藤、罗勒;具有减弱GLU诱导PC12细胞损伤的药物为:虎爪豆、诃子、凤仙花、仙人掌、芦笋、甘草;诃子在3种模型中均表现出较强的细胞损伤保护作用。本研究结果为神经退行性疾病药物的开发和选择提供了初步的实验依据。
This study aimed to investigate the cytoprotection of 20 extracts from 13 Pakistan medicinal plants on PC12 cells injured by H2O2 and glutamate (GLU). Different concentrations of H2O2 and GLU were used to establish the toxicity models on PC12 cells. Cell viability was determined by MTY assay. The results showed that the extracts of Mucuna pruriens L and Terminalia chebula had significantly proliferative effects on PC12 cells (P〈0.05). Additionally, extracts of Lawsonia alba, Opuntia dillenii Haw., Celastrus paniculata, Terminalia chebula showed markedly cytoprotection against H2O2-induced cell death (P〈0.05). Similar results were observed in GLU injured model, which showed that extracts of Lawsonia alba, Opuntia dillenii Haw., Mucuna pruriens L. and Terminalia chebula attenuated GLU-induced toxicity obviously (P〈0.05). Among all extracts, only Terminalia chebula had cytoprotection against both H2O2 and GLU induced neurotoxicity with proliferative effect on PC12 cells. It is concluded that extracts of Terminalia chebula, Ocimum basilicum, Mucuna pruriens L. have promoting effect on the proliferation of PC12 cells. Terminalia chebula, Lawsonia alba, Opuntia dillenii Haw., Asparagus officinalis, Celastrus paniculata, Ocimum basilicum extracts possess significant antioxidative activities. And the extracts from Mucuna pruriens L, Terminalia chebula, Lawsonia alba, Opuntia dillenii Haw., Asparagus offwinalis, Glycyrrhiza glabra L. can decrease damages induced by GLU in PC12 cells. The results suggested that Terminalia chebula had better cytoprotection against injured models on PC12 ceils. Results of this study provide a preliminary experimental basis of screening and developing new agents for neurodegenerative diseases.
