摘要
目的探讨高糖环境下人肾小球系膜细胞中p38丝裂原活化蛋白激酶/cAMP反应元件结合蛋白(p38MAPK/CREB)通路的活性及细胞外基质成分纤维黏连蛋白(FN)的变化。方法以5.6mmol/L葡萄糖培养基培养肾小球系膜细胞为对照组,观察高糖(30mmol/L)培养12、24、48小时。肾小球系膜细胞p38MAPK/CREB活性和FNmRNA、FN的变化。结果与对照组比较,高糖组各时间p-p38MAPK、P—CREB的表达均上升,并随着时间的延长,其表达逐渐升高;FN的表达升高,并且随着时间的延长,其表达进一步升高。结论高糖环境下p38MAPK/CREB通路被激活,FN表达增加,p38MAPK/CREB通路参与了细胞外基质重构的过程。
Objective To investigate the activity of p38MAPK/CREB pathway and the change of fibronectin(FN) in human mesangial cell(HMCs) cultured under high glucose condition. Methods HMCs which were treated with 5.6 mmol/L glucose medium can be thinked as Normal control group, we can observe the change of the activity of p38MAPK, CREB, protein of FN, and mRNA of FN; Western Blot was used to assess the expression of protein of p-p38MAPK and p-CREB;Semi-quantitative RT-PCR was used to assess the expression of FNmRNA;Elisa was used to assess the expression of FNo Results Compare with the Normal control group,expression of p-p38MAPK and p-CREB were significantly enhanced and with the prolongation of time,the expression were higher and higher;expression of FN was also increased, and with the prolongation of time, the expression were higher and higher. Conclusions p38MAPK/CREB pathway was activitied under high glucose condition, and the expression of FN increase, so p38MAPK/CREB pathway may take part in the reconstitution of extracellular matrix(ECM).
