胎盘组织中HIF-1α的表达与血清sFlt-1,sEng水平在子痫前期的作用研究 被引量:3

摘要 目的探讨胎盘组织中缺氧诱导因子-1α(HIF—1α)的表达和血清中可溶性Endoglin(sEng)、可溶性血管内皮细胞生长因子受体-1(sFlt-1)水平在子痫前期发病机制中的作用。方法采用免疫组化SABC法检测20例正常晚期妊娠孕妇(对照组)和35例子痈前期患者(予痫前期组,其中轻度16例、重度19例)胎盘组织中HIF—1α的表达;采用酶联免疫吸附实验的方法检测其血清中sEng和sFh—1水平。结果①轻、重度子痈前期组中HIF-1α在合体滋养细胞的表达显著高于对照组(H=19.3,P〈0.05;H=23.1,P〈O.01)。②轻、重度子痫前期组血清sEng,sFlt-1浓度分别为(4.39±0.48)ng/m1,(5.61土0.66)ng/ml;(22.69±5.38)Pg/L,(30.15±5.43)vg/L,均显著高于对照组(1.71±0.27)ng/ml,(6.61±2.43)pg/L,差异具有统计学意义(t=21.25,P〈0.01;t=24.47,P〈0.01);(t=11.96,P〈0.01;t=17.63,P〈0.01)。重度组高于轻度组0=6.17,P〈0.01;t=4.06,P〈0.01)。③子痫前期胎盘组织中HIF-1α表达与血清sEng,sFlt-1水平呈正相关(r=0.76,P〈0.01;r=0.87,P〈0.01)。结论HIF-1α和sEng,sFlt-1共同参与子痫前期的病理生理过程,并与病情严重程度有一定关系。 Objective To investigate the expression of hypoxia-inducible factors-1α in (HIF-1α) placenta and serum soluble endoglin,soluble fins-like tyrosine kinase receptor-1 in maternal serum from normal pregnant women and preeclampsia,in order to probe the possible mechanism of preeclampsia. Methods The expression of HIF-1α was detected by immunohis- tochemistry. The levels of sEng and sFh-1 in serum were detected by enzyme linked immunosorbent assay. Results (1)The expressions of HIF-1α in placental villous syncytiontrophoblast of preeclampsia group were significantly higher than that of control group (H = 19.3,P〈0.05 ; H = 23.1 ,P〈0. 01). (2)The level of sEng,sFh-1 in serum of the the mild preeclampsia group (4. 39± 0. 48)ng/ml, (22.69±5.38) μg/L and the sever group (5.61±0. 66) ng/ml, (0. 15±5.43)μg/L were significantly higher than that of control group (1. 71±0. 27) ng/ml, (6. 61±2.43) μg/L; (t=21.25,P〈0. 01 ;t=24. 47,P〈 0. 01) and (t=11. 96,P〈0. 01 ;t= 17.63,P〈0. 01). The sever group was much higher than the mild group (t=6. 17,P〈 0. 01 ;t= 4.06,P〈0. 01). (3)There were significant correlation between he expressions of HIF-1α in placental and the level of sEng ,sFh-1 in serum of mild preeclampsia group and sever group (r= 0. 76 ,P〈0. 01 ;r=0. 87 ,P〈0. 01). Conclusion HIF-1α,sEng and sFh-1 not only paticipated in pathogenic process of preeclampsia but also related to its degrees.
