奥美沙坦对心肌梗死后心力衰竭大鼠内皮素1和肾上腺髓质素水平的影响 被引量:4

Effects of olmesartan therapy on plasma levels of ET-1 and ADM in rats with heart failure induced by myocardial infarction

摘要 目的探讨奥美沙坦对心肌梗死后心力衰竭大鼠血浆内皮素1(endothelin-1,ET-1)、肾上腺髓质素(adrenomedullin,ADM)水平的影响。方法 Wistar大鼠38只,随机分为对照组10只、假手术组10只、模型组9只、奥美沙坦组9只。采用结扎大鼠冠状动脉左前降支的方法制作心肌梗死后心力衰竭模型,灌胃法给药8周。超声心动图检测用药前后大鼠心功能,放射免疫法测定血浆ET-1、ADM水平。结果用药前,对照组与假手术组大鼠LVEF、ET-1、ADM水平差异无统计学意义(P>0.05);与假手术组比较,模型组和奥美沙坦组大鼠LVEF明显降低,差异有统计学意义(P<0.01)。用药后,与假手术组比较,模型组大鼠ET 1、ADM水平明显升高,差异有统计学意义(P<0.05,P<0.01);与模型组比较,奥美沙坦组大鼠LVEF明显改善,ET-1、ADM水平明显降低,差异有统计学意义(P<0.05,P<0.01)。结论应用奥美沙坦治疗可以改善心肌梗死后心力衰竭大鼠心功能,降低心力衰竭时升高的血浆ET-1、ADM水平,抑制慢性心力衰竭时肾素-血管紧张素系统以外的神经内分泌系统激活,对慢性心力衰竭心脏起到保护作用。 Objective To investigate the effect of olmesartan on the plasma levels of endothelin-1 (ET-1) and adrenomedullin(ADM) in rats with chronic heart failure(CHF) induced by myocardial infarction. Methods Thirty-eight Wistar rats were randomly divided into four groups:control group(10 rats), sham operation group(10 rats), model group (9 rats) and olmesartan group (9 rats). Heart function was examind by ultrasonogram,plasma ET-1 and ADM levels were meas- ured by radioimmunoassay. Results Before therapy, no difference between control group and sham operation group was observed. Compared with sham operation group,the ejection fraction of left ventricle(LVEF) of model group and olmesartan group was significantly decreased (P 〈0.01). After therapy,compared with sham operation group,plasma levels of ET-1 and ADM of model group were markedly increased (P 〈 0.05 ,P 〈 0.01). Compared with model group,LVEF of olmesartan group was significantly improved, plasma levels of ET-1 and ADM of olmesartan group were markedly decreased (P 〈0.05, P〈 0.01). Conclusions Heart function was improved and levels of ET-1 and ADM were decreased when olmesartan as a therapy for rats with CHF induced by myocardial infarction.
