大肠癌中抑癌基因WWOX及相关基因Bax、E2F1的表达及意义

摘要 目的:探讨WWOX(WW domain containing oxidoreductase)基因的表达与大肠癌生物学行为及Bax(Bcl-2-associated X protein)、E2F1(E2F transcription factor 1)表达的相关性。方法:应用免疫组化Envision二步法检测58例大肠癌中WWOX、Bax及E2F1的表达,并以20例大肠粘膜炎症为对照。结果:(1)WWOX、Bax在大肠癌中的阳性率分别为25.86%、58.62%,显著低于大肠粘膜炎症的65%、85%,差异有统计学意义。(P〈0.01,P〈0.05);E2F1在癌组织中的阳性率为63.79%,显著高于炎症组的35%,差异有统计学意义(P〈0.05)。(2)E2F1在大肠癌中的表达与浸润深度有关(P〈0.05);WWOX、Bax在大肠癌中的表达与患者的预后相关,差异有统计学意义(P〈0.01,P〈0.05),且WWOX的表达在癌组织的浸润深度间差异有统计学意义(P〈0.05)。(3)WWOX与E2F1的表达无相关性(x^2=2.301,P〉0.05),而与Bax的表达有关(x^2=6.561,P〈0.05)。结论:WWOX、Bax及E2F1参与调控大肠癌的发生、发展,且WWOX与Bax协同作用,在大肠癌的预后中扮演了重要角色。 Objective: To study the relationship between the expression of WWOX (WW domain containing oxidoreductase) and its biological behavior, as well as its correlation with the expressions of Bax( Bcl-2-associated X protein) and E2F1 (E2F transcription factor 1 ) in colorectal carcinoma. Methods : The expressions of WWOX, Bax and E2F1 were detected by Envision immunohistochemical staining in 58 cases of colorectal carcinoma tissues, and compared with those in 20 cases of colorectal mucosal inflammations. Results : ( 1 ) The positive rates of WWOX, Bax were 25.86% , 58.62% respectively in colorectal carcinoma, which were much lower than those 65% , 85% in colorectal mucosal inflammations. The difference was significant between the two groups ( P 〈 0. 01, P 〈 0. 05 ). The positive rate of E2F1 was 63.79% in colorectal carcinomas, which was much higher than that in colorectal mucosal inflammations(35% ). The difference was significant( P 〈 0. 05 ). (2)The expression of E2F1 in colorectal carcinoma tissues correlated with the invasion depth( P 〈 0. 05 ). The expressions of WWOX and Bax in colorectal carcinomas tissue correlated with the prognosis( P 〈0. 01 ,P 〈 0. 05 ). In the coloretal carcinoma tissues with different invasion depth, the expressions of WWOX were significantly different( P 〈 0. 05 ). ( 3 ) Expression of WWOX was positively correlated with Bax (x^2 = 6. 561, P 〈 0. 05 ), but not with E2F1 (X2 = 2. 301, P 〉 0. 05 ). Conclusion : WWOX, Bax and E2F1 correlate with occurrence and development of colorectal carcinoma. WWOX and Bax play a key role in the prognosis of colorectal carcinoma.
