化浊解毒活血方对急性心肌缺血大鼠心肌酶和血液流变学的影响

Effect of Huazhuo Jiedu Huoxue Recipe on cardiac enzyme and hemorheology in rats with acute myocardial ischemia

摘要 目的观察化浊解毒活血方对急性心肌缺血大鼠心肌酶和血液流变学的影响。方法将50只W istar雄性大鼠随机分为5组,即空白对照组、模型组、消心痛对照组及化浊解毒活血方高、低剂量组(以下分别简称中药高、低剂量组),每组10只。空白对照组和模型组分别予0.9%氯化钠注射液10 mL/kg灌胃,消心痛对照组予消心痛悬浮液(由消心痛2.5 mg/kg加入0.9%氯化钠注射液10 mL配制而成)10 mL/kg灌胃,中药高、低剂量组分别予化浊解毒活血方高剂量(含生药4.70 g/mL)、低剂量(含生药2.35 g/mL)10 mL/kg灌胃,均每日1次,连续10 d。第10 d末次给药后1 h,除空白对照组外,其余各组采用冠状动脉左前降支结扎法制作心肌缺血模型,空白对照组只手术,不结扎。造模后继续给药2 d。取各组大鼠腹主动脉血,测定各组血液流变学指标全血黏度(高切)、全血黏度(低切)、血浆黏度及红细胞比容。然后断头处死各组大鼠,取动脉血,测定肌酸磷酸激酶(CPK)、天冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)。结果模型组及各药物组各血液流变指标与空白对照组比较差异均有统计学(P<0.05),造模成功。各药物组全血黏度(高切)、全血黏度(低切)与模型组比较均明显下降(P<0.05)。中药高剂量组血浆黏度、红细胞比容及低剂量组红细胞比容均低于模型组(P<0.05)。中药高剂量组各血流变指标与消心痛对照组比较差异均有统计学意义(P<0.05)。模型组及各药物组CPK、AST、LDH与空白对照组比较均明显升高(P<0.05),造模成功。且各药物组CPK、LDH及中药高、低剂量组AST与模型组比较均明显下降(P<0.05)。中药高、低剂量组CPK、AST、LDH与消心痛对照组比较差异无统计学意义(P>0.05),且中药高、低剂量组间比较差异亦无统计学意义(P>0.05)。结论化浊解毒活血方能降低CPK、AST、LDH活性,改善心肌缺血大鼠血液流变学指标,对缺血心肌具有保护作用,其作用机制可能为扩张冠状动脉,改善心肌内血液循环,增加冠脉流量,增加心肌供氧。 Objective To study the effect of Huazhuo Jiedu Huoxue Recipe on cardiac enzyme and hemorheology in rats with acute myocardial ischemia.Methods Fifty Wistar male rats were randomly divided into five groups,namely control group,model group,isosorbide dinitrate group and Huazhuo Jiedu Huoxue Recipe high-and low-dose group(hereinafter referred to as the high-and low-dose group),n=10 in each group.The control group and model group were to be 10 mL/kg 0.9% sodium chloride injection,gavage.Isosorbide dinitrate group orally received suspension of 10 mL/kg isosorbide dinitrate to(from 2.5 mg/kg of isosorbide dinitrate added 10 mL 0.9% sodium chloride injection formulated).High-and low-dose group were to Huazhuo Jiedu Huoxue Recipe high dose(containing crude drug 4.70 g/mL),low-dose(containing crude drug 2.35 g/mL) 10 mL/kg orally once a day,for continuous 10 d.The first 10 d after the last administration 1 h,in addition to the control group,the rest of the group model of myocardial ischemia were produced with left anterior descending coronary artery ligation,only the surgical,and no ligation in control group.Administration was continued to 2 d after modeling.Blood from abdominal aorta of rats was taken to measure whole blood viscosity and blood hemorheology in each group,whole blood viscosity(high shear,low shear),plasma viscosity and hematocrit.Then the rats were decapitated,taking blood to measure creatine phosphokinase(CPK),aspartate aminotransferase(AST),lactate dehydrogenase(LDH).Results Hemorheology indexes and CPK,AST,LDH in model group and each treatment group were differ from those in control group(P0.05),modeling was success.There were significant differences between each treatment group and model group on whole blood viscosity(high shear),whole blood viscosity(low shear),CPK and LDH(P0.05).There were significant differences between high-dose group and model group on the plasma viscosity and hematocrit(P0.05).There was significant difference between low-dose group and model group on hematocrit(P0.05).There were significant differences between high-dose group and isosorbide dinitrate group on hemorheology indexes(P0.05).Conclusion Huazhuo Jiedu Huoxue Recipe can reduce the enzymes CPK,AST,LDH activity,improve myocardial ischemia hemorheology,has a protective effect on myocardial ischemia,its mechanism may be the expansion of the coronary arteries,improving blood circulation within the heart,increasing coronary blood flow,increasing myocardial oxygen supply.
