摘要
利用AFLP技术,对鄂柑一号椪柑(Citrus reticulata Blanc.cv.ponkan)的3个少核芽变及其母株进行分子鉴定。筛选了64对引物,有60对引物可扩增出清晰的条带,共获得2 281条清晰的扩增谱带,其中有9对引物可扩增出明显多态性条带,多态性位点共15个,占总数的0.66%。结果表明,鄂柑一号椪柑的少核芽变由遗传物质改变引起,通过AFLP技术可以有效鉴定。
Three few-seed bud mutations and their mother plant 'E-Gan No.1' ponkan were identificated by AFLP technique.64 selective primer combinations were tested and 2 281 clear bands were abtained from 60-pair-primer,from which 9-pair-primer were selected for further reseach.And 15 polymorphic bands were dectected out of 2281 total clear bands;the polymorphic rate was 0.66%.This results indicated that there were some molecular level differentiation between E-gan No.1 and the three few-seed bud mutations;and the differentiation could be identificated by AFLP.
