桃仁红花煎对血管性痴呆大鼠学习记忆能力的影响 被引量:11

Effects of Taorenhonghua Decoction on Learning and Memory in Rats with Vascular Dementia

摘要 目的探讨桃仁红花煎对慢性全脑缺血诱导的血管性痴呆大鼠的作用。方法大鼠双侧颈总动脉永久结扎建立慢性不完全性全脑缺血模型,实验组每日灌胃桃仁红花煎,假手术组和模型组每天分别给予等体积0.9%氯化钠溶液。30 d后进行水迷宫行为学检测。结果与假手术组找到平台的潜伏期(20.1±2.5)s比较,模型组大鼠找到平台的潜伏期明显延长([33.4±3.9)s,P<0.05;]与模型组比较,实验组大鼠找到平台的潜伏期显著缩短([24.5±3.1)s,P<0.05]。模型组在平台所在象限的探索时间为(37.5±3.3)s,较假手术组在平台所在象限的探索时间([20.3±4.0)s]明显延长(P<0.05),实验组在平台所在象限的探索时间为(29.1±3.6)s,较模型组明显缩短(P<0.05)。海马组织丙二醛(MDA)、超氧化物歧化酶(SOD)和过氧化氢酶(catalase,CAT)检测显示,模型组MDA水平较假手术组明显升高,而SOD和CAT活性明显降低,实验组有效地逆转了上述改变。形态学检测显示桃仁红花煎能有效减轻慢性全脑缺血诱导的海马组织神经元的损害。结论桃仁红花煎可有效改善慢性全脑缺血诱导的血管性痴呆大鼠的学习和记忆能力,这种效应可能与其抗氧化作用有关。 Objective To investigate effects of taorenhonghua decoction on rats with vascular dementia induced by chronic global isehemia. Methods Permanent occlusion of the bilateral carotid arteries was established in rats. The treatment group was daily orally administered with taorenhonghua decoction after surgery. Rats in sham and model groups were treated with equal-volume of saline daily. Morris water maze test was conducted on day 30 post-surgery. Results Rats in model group [ (33.4±3.9) s ] consistently took longer to find the hidden platform than those in sham group (20.1±2.5) s (P〈0.05). The prolonged escape latency in treatment group ( 24. 5 ± 3. 1 ) s was significantly shortened by long-term administration of taorenhonghua decoction (P〈0.05). In the spatial probe trials, the rats in model group spent more time ( 37.5± 3.3 ) s on searching the target quadrant where the platform was located than the sham group (20.3±4.0) s ( P〈0.05 ). The rats in treatment group spent less time (29.1 ±3.6) s in searching the platform in the target quadrant than the model group did, and the significant difference was observed between model and treatment groups ( P〈0.05 ). Compared with sham-operated group, MDA levels were significantly increased, while SOD and catalase activities were significantly decreased in model group, while which were reversed by long-term administration of taorenhonghua deeoetion in treatment group. HE staining showed neuronal loss, shrinkage and marked vacuolar changes in the hippoeampus of model group, which was significantly attenuated by taorenhonghua decoction. Conclusion Taorenhonghua decoction can improve the study and memory ability of rats with vaseular dementia induced by chronic global isehemia, which may partly be related to its antioxidant actions.
