摘要 目的:为满足人们栽培对种苗的需要,并保护野生资源。方法:以地丁草不同外植体为材料,进行了愈伤组织诱导与分化、不定芽生根和试管苗生根继代培养研究,建立起地丁草的无性系。结果:MS+NH4H2PO4 20mg·L-1+BA0.2mg·L-1+IBA0.1mg·L-1+NAA0.8~1.0mg·L-1是愈伤组织继代扩增培养的理想培养基;MS+AgN030.5mg·L-1+BA0.8mg·L-1+NAA0.1mg·L-1是愈伤组织继代扩增培养的愈伤组织分化理想培养基。MS+AgNO。0.5mg·L-1+BA0.8mg·L-1+NAA0.1mg·L-1是愈伤组织分化理想培养基。1/2MS+IAA0.2mg·L-1-0.4mg·L-1是试管苗生根培养的理想培养基。1/2MS+IAA0.3mg·L-1是试管苗生根继代培养的理想培养基。移植到草地上的试管苗保持了野生地丁草的所有生物性,当年鲜草产量增加32%。结论:建立起地丁草的无性系。 Objective : To meet the need for cultivation of germchits and protect wild resources. Methods : Dif- ferent explants from Corydalis bungeana were cultured in vitro to investigate callus inducement and differentia- tion, formation of adventitious roots,multiplication of adventitious roots and the clones of Corydalis bungeana was established. Results: The ideal medium for callus multiplication was MS+NH4H2PO4 20mg·L-1+BA0.2mg·L-1+IBA0.1mg·L-1+NAA0.8~1.0mg·L-1;the medium of MS+AgN030.5mg·L-1+BA0.8mg·L-1+NAA0.1mg·L-1 was suitable for callus differentiation; 1/2MS+IAA0.2mg·L-1-0.4mg·L-1 was the best rooting medium for the tube seedlings, and the multiplication of adventitious root ideal medium was 1/2MS + IAA0.3mg·L-1. The tube seedlings retained all biological characteristics of the escape Corydalis bungeana af- ter transplanted to grassplot. The year yield of fresh grass was increased by 32%. Conclusion:The clone of Corydalis bungeana is established.
