摘要
目的:研究bFGF和rhG-CSF单独和联合应用对缺血性脑卒中模型鼠神经功能和神经分化特异性抗体CD34,MAP-2表达的影响。方法:线栓法造模,bFGF组脑区注射bFGF,动员剂组rhG-CSF,联合组同做。分别于干预不同时段神经功能评价及脑区切片免疫荧光检测CD34,MAP-2表达。结果:处理早期,联用组及rhG-CSF组神经功能改善,CD34阳性细胞数较多;处理10d后,bFGF组神经功能改善明显,MAP-2阳性细胞数增多;联用组的CD34及MAP-2阳性细胞数均多于其他组。结论:rhG-CSF在使用10d内明显促进神经功能改善及CD34阳性细胞数量增加;bFGF作用15d达高峰,明显促进神经功能改善及MAP-2抗体的表达,显示出病灶区形成突触的有功能神经元的数量的增加;联合组对病灶区神经结构及功能都显示两因素的协同作用。
Objective: To investigate the effects of bFGF and rhG--CSF in combination or alone on neural function and CD34 and MAP--2 express of encephalic specific antibodies in cerebral arterial thrombosis rat model. Methods: We made the cerebral arterial thrombosis rat model by using line block mode by injecting bFGF into encephalic region in bFGF group and injecting rhG--CSF into cervical region in mobilizing agent group and both in the combined group . Then we evaluated neural function at different times and detected CD34 and MAP--2 antibodies by immunofluorescence. Results: The rhG--CSF group and the combined group showed the improvement of neural function and had more CD34 positive cells after 2 weeks, and the bFGF group showed noted improvement and had more MAP--2 positive cells after10 days. The combined group had more CD34 and MAP--2 positive cells than the other two groups. Conclusion: 10 days after rhG--CSF is used, the neural function is significantly improved with increased CD34 positive cells. The bFGF effect reaches a peak at 15th day, with significant improvement of neural function and increased MAP--2 positive cells. The two factors show a synergistic effect on the neural structure and function in the combined group.
