摘要
为建立一种简便、快捷、灵敏的硫酸软骨素(CS)测定方法,研究天青Ⅰ染色剂直接测定CS含量的分光光度法,二者能特异性结合成蓝紫色复合物,该复合物在特定波长处有最大光吸收,通过测定吸光度值可测CS含量。结果天青Ⅰ-CS复合物在波长547 nm处有最大光吸收,且在10 mg/L^100 mg/L范围内,具有良好的正相关线性关系,回归方程是y=0.040 4x+0.248 3,R2=0.993 7;该测定方法在5 h内稳定性、重现性测定结果P>0.05,说明数据可靠,加样平均回收率达94.34%,是一种简便、快捷、灵敏的测定方法。
Aimed to establish a simple,rapid and sensitive method to determine the content of chondronitin sulfate,the research study on directly spectrophotometric determination about the content of chondronitin sulfate with Azure Ⅰ.They could form a specific-binding blue-purple complex that was Azure Ⅰ-chondronitin sulfate.At the characteristic wavelength,this complex had the max absorption,and the content of chondronitin sulfate could be determined by absorption value.The result was that the complex had the max absorption at 547 nm.A good positive linear relationship was shown in the mass concentration range of chondronitin sulfate 10 mg/L-100 mg/L.The regression equation was y=0.040 4x+0.248 3,R2=0.993 7.With this method,the result of the stability in 5 h and the repeatability was well and reliable that P〉0.05.The recovery average rate of added samples was 94.34 %.It was a simple,rapid and sensitive determination method.
