摘要
以甘蔗(FN95-1702)为材料,首次克隆得到甘蔗UGPase DNA片段,测序结果表明该基因全长约5.3 kb,包含21个外显子,20个内含子。开放读码框(ORF)共编码476个氨基酸。通过接头连接PCR方法克隆得到约0.8 kb的该基因5′侧翼序列,并对得到的序列进行基础启动子区及调控元件分析,预测在该序列上存在TATA-box、CAAT-box等特征元件,此外还预测到大量与光响应有关的的顺式作用元件和一些与组织发育相关的元件。
A DNA encoding UGPase was isolated from the leaf of sugarcane, The sequence consists of 5.3 kb, has twenty one exons and twenty introns. The open reading frame encodes a protein of 476 amino acids. The 5' flanking sequence of UGPase was cloned by Adaptor-ligation PCR, then analyzed the foundation promoter and the controlling element of regulatory sequence, some characteristic elements (TATA-box, CAAT-box), light responsive elements and some elements related tissue development were calculated.
