摘要 目的观察过氧化氢(H2O2)对老年大鼠血管平滑肌细胞(VSMC)增殖的影响。方法主动脉贴块法分别培养3月龄和18月龄大鼠的VSMC,分为3月龄组和18月龄组,各5只大鼠,比较两组VSMC在不同浓度H2O2(0、10、20、30、40、50、60、80和100μmol/L)作用6h下的增殖情况。采用四甲基偶氮唑盐微量酶反应经色法测定细胞的增殖能力。结果两组未给予H2O2时VSMC增殖能力无差异。与3月龄组比较,18月龄组大鼠VSMC在较低浓度H2O2(10~60μmol/L)作用时表现为更强的增殖能力,而在较高浓度H2O2(80~100μmol/L)作用下VSMC增殖能力降低更明显。结论老年大鼠VSMC对氧化应激损伤的反应明显强于成年大鼠VSMC,这可能是老年期易发动脉粥样硬化的机制之一。 Objective To observe effects of hydroperoxide on the proliferation of vascular smooth muscle cells (VSMCs)obtained from different age strata rats. Methods The VSMCs from the aorta of 18 months old rats and 3 months old rats were cultured using explant method,and age related proliferation changes of sensitivity of VSMCs under serial concentration of H202(0,10,20,30, 40,50,60,80 and 100 μmol/L) were observed using MTr method. Results Ability of proliferation of VSMCs without H202 in two groups was not different. Compared with 3 months old rats,the VSMCs proliferation was higher in 18 months old rats incubation with low concentration H202(10-60μmol/L). While incubation with high concentration H2O2(80- 100 μmol/L), the VSMCs proliferation was lower and the cells were more liable to occur necrosis and apoptosis in 18 months old rats. Conclusions Damage of VSMCs proliferation to ROS in 18 months old rats is more severe than that in young rats,and this maybe one of the mechanisms that ageing is easy to suffer from atherosclerosis.
