基于40种中药多糖HPLC图谱的寒热药性Fisher分析 被引量:4

摘要 目的:研究40种中药多糖成分与中药寒热药性的相关性。方法:采用水提醇沉法提取中药多糖,用三氟乙酸彻底水解成单糖,以1-苯基-3-甲基-5-吡唑啉酮进行衍生化,高效液相色谱法(HPLC)测定多糖的单糖组成,利用Fisher方法分析实验结果。结果:Fisher法建立判别函数。判别函数常数项为4.35146,判别函数为Y=4.35146+系数×X保留时间。计算判别函数得分,得分为正值判为寒性中药,为负值判为热性中药。判别结果显示,只有寒性中药甘遂和天冬判别错误,其余均判别正确,组内回代一致率为95%。结论:中药寒热药性与多糖成分存在相关性,多糖成分是表征中药寒热药性的影响因素之一。 Objective: To study the correlation between polysaccharides and cold-heat nature of 40 kinds of herbs.Methods: The polysaccharides were extracted by water and precipitated by ethanol,then completely hydrolyzed into monosaccharides by using 2 mol/L trifluoroacetic acid.The monosaccharide was derivatized with 1-phenyl-3-methyl-5-pyrazolone and separated by RP-HPLC using a developed gradient elution process,and monitored by ultraviolet detector at 250nm.Finally the results of the HPLC of monosaccharides composition of polysaccharides were analyzed by Fisher method.Results: From the results,it reveals that the monosaccharide standards separated well.Determining the HPLC of 40 herbs by the same conditions of the standards,it showed the 20 cold herbs were more than the 20 heat herbs in the numbers of the peaks which represented the kinds of the monosaccharides as well as the peak areas which were the representative of the contents.The Fisher method established a discriminant function.The results confirmed that there were many differences between the cold-heat nature.The discriminant result of the Fisher analysis was 95%.Conclusion: The results demonstrated that there was a correlation between cold-heat nature and polysaccharides of 40 kinds of herbs.The Fisher analysis could distinguish the cold-heat nature.In addition,it also concluded that the polysaccharide was one of the material basis of cold-heat nature.
