益气养阴解毒方对K562/A02裸鼠移植瘤逆转作用研究

Study of effect and reversal mechanism of YiQiYangYinJieDu Prescription on transplantation tumor induced by K562/A02 cell in nude mice

目的:研究益气养阴解毒方对K562/A02多药耐药移植瘤的逆转作用。方法:建立K562/A02裸鼠移植瘤多药耐药模型;选用维拉帕米作阳性对照,用流式细胞仪(FCM)检测各组移植瘤耐药细胞P糖蛋白(P-gp)与bcl-2的表达情况和移植瘤细胞内注射用盐酸多柔比星(ADM)蓄积浓度及移植瘤耐药细胞的凋亡情况。结果:移植瘤P-gp表达与维拉帕米组相比,生理盐水组与中药小剂量组有差异(P<0.05);bcl-2的表达与维拉帕米组相比,中药大剂量组有差异(P<0.05);益气养阴解毒方能使移植瘤耐药细胞内ADM的浓度升高。结论:益气养阴解毒方具有部份逆转白血病K562/A02移植瘤多药耐药性的作用,其逆转移植瘤裸鼠多药耐药性的机制不是通过下调肿瘤细胞膜上P-gp的表达实现的,而与降低bcl-2的表达、促进耐药细胞凋亡有关。

Objective：To study the effect and reversal mechanism of Chinese compound recipe on transplantation tumor induced by K562/A02 cell.Methods： Establish the model of transplantation tumor of K562/A02 in Nude mice;set the positive contract group of Verapamil,FCM was used to investigate the changes of expression of P-gp and bcl-2,the accumulation of intracellular ADM,and the apoptosis of K562/A02 cell invivo after different drug treatment.Results：Expression of P-gp： Compared with Verapamil group,there were significant differences in the group of normal saline and the group of small dosage of Chinese medicine（P〈0.05）;The expression of bcl-2： Compared with Verapamil group,there was significant difference in Chinese medicine of large dosage（P〈0.05）.Conclusion： YiQiYangYinJieDu Prescription could increase the accumulance of intracellular adria mycine and effectively reverse the multidrug resistance of transplantation tumor,the mechanism of reversal may be not related with inhibition of P-gp expression but weakening the expression of bcl-2 protein and promotion the K562/A02 apoptosis.
