摘要
目的研究尼古丁损伤雄性小鼠生殖功能的可能机制。方法选用8周龄的昆明小鼠24只,随机分成两组,实验组皮下注射尼古丁(4.286 mg.kg-1.d-1)8周,对照组皮下注射等量0.9%氯化钠注射液。8周后,检测小鼠附睾中精子数目及精子活力、睾丸和血清中睾酮含量、睾丸中一氧化氮(NO)含量和相关诱导型NO合酶的蛋白表达情况。结果实验组较对照组精子数目减少(P<0.05),精子活力下降(P<0.05),睾丸和血清中睾酮含量下降(P<0.05),睾丸中NO含量增加(P<0.05),诱导型NO合酶表达增加(P<0.05)。结论在尼古丁的刺激下,睾丸中NO的含量增加,可能是导致生殖功能损伤的机制之一。
Objective To study the contribution of nitric oxide(NO)to nicotine-induced reproductive injury in male mice.Methods 24 males Ku-Ming mice with eight weeks old were randomly divided into normal saline group,nicotine treated group(subcutaneous injection with nicotine 4.286mg·kg-1·d-1)for 8 weeks.The number and motility of epididymis sperm,the concentration of testosterone in testis and serum,the level of nitric oxide and NO synthase in testis were detected.Results Compared with normal saline group,the number and motility of sperm decreased,and so did the concentration of testosterone in testis and serum in nicotine-treated group(P〈0.05).Whereas,the concentration of NO and the expression of inducible NO synthase increased in nicotine-treated group(P〈0.05).Conclusion The augmentation of nitric oxide level may contribute to the nicotine-induced reproductive injury in male mice.
