储藏湿度对水杉种子同工酶的影响 被引量:2

Influence of different humidity storage conditions on enzymes of Metasequoia glyptostroboides seeds germination

摘要 以不同储藏湿度下的水杉种子为材料,采用非变性聚丙烯酰胺凝胶电泳技术对水杉种子中的过氧化物酶(POD)、酯酶(EST)、超氧化物歧化酶(SOD)、淀粉酶(AMY)、ATP酶(ATPase)等5种同工酶进行分析。结果表明:4℃、50%湿度沙藏是最适合水杉种子储藏的条件;当储藏湿度不同时,POD、EST、SOD、ATPase的差异显著,其中POD、SOD和ATPase随湿度的增加,其含量均先升高再降低,当湿度为50%时达到最大;EST在20%和50%湿度条件下,酶带数和含量均较好;储藏湿度对AMY无显著影响。 Non-denaturing polyacrylamide gel electrophoresis was used as a tool to analyze isozyme patterns such as peroxidase(POD),esterase(EST),amylase(AMY)and superoxide dismutase(SOD) of seeds of Metasequoia glyptostroboides in different humidity storage conditions.The results showed that 4℃,50% humidity Sand was the most suitable storage conditions for Metasequoia seed.In different humidity storage conditions,POD,EST,SOD and ATPase isozymes were significantly difference.With the humidity increasesing,the content of POD,SOD and ATPase isozymes increased and then decreased,they reached maximum when the humidity was 50%.In 20% and 50% humidity conditions,enzyme levels of EST isoenzyme were better.But with humidity changing,the difference of the AMY isozymes was not obvious.
