摘要
利用根癌农杆菌介导法将携带有O型和A型口蹄疫抗原决定簇融合基因O21-O14-A21的双元表达载体对拟南芥外植体进行遗传转化,并探讨了不同侵染时间和菌液浓度对农杆菌遗传转化效率的影响。通过潮霉素筛选,共获得522块抗性愈伤组织,经再生培养得到36棵再生苗,对部分再生植株进行PCR检测,结果表明目的基因已整合到拟南芥基因组中。
The construct carrying O and A types of FMDV antigenic determinant fused gene O21-O14-A21 was transformed into the whole plant of Arabidopsis thaliana by Agrobacterium-mediated transformation method.The impact of transformation efficiency by the infection time and Agrobacterium concentration were investigated.522 hygromycin-resistant callus were obtained by screening on the medium containing antibiotic,and 36 regenerative plantlets were acquired after regenerative culture.Confimation of the transgenic plants by PCR analysis demonstrated that target gene has been integrated into genome of the transformed plants.
