复方绞股蓝胶囊对口腔鳞癌CAL27细胞增殖和表达的影响(英文)

摘要 目的研究复方绞股蓝胶囊(CoFG)对口腔鳞癌CAL27细胞MMP-2,9和Bax、Bcl-2表达的影响。方法采用MTT法检测CoFG对口腔鳞癌CAL27细胞增殖抑制的作用;qPCR法和Western blot分别检测CoFG对口腔鳞癌CAL27细胞MMP-2,9和Bax、Bcl-2 mRNA及蛋白表达的影响。结果 CoFG组和非CoFG组在细胞增殖抑制方面差异有统计学意义;CoFG下调MMP-2、Bcl-2 mRNA及蛋白表达;CoFG上调MMP-9、Bax mRNA及蛋白表达。结论 MMP-2,9和Bax、Bcl-2在口腔鳞癌的发生发展中起一定作用,CoFG可能对口腔鳞癌的治疗具有一定的潜在作用。 AIM To investigate the influence of compound fiveleaf gynostemma capsules(CoFG) on the expression of Matrix metalloproteinases(MMPs)-2,9 and Bax,Bcl-2 in the CAL27 cells of Oral squamous cell carcinoma(OSCC). METHODS MTT assay was used to examine the effect of CoFG on the cytostasis of CAL27 cells of OSCC.The expression of mRNA and protein of MMP-2,9 and Bax,Bcl-2 in the CAL27 cells of OSCC treated by CoFG were assayed by qPCR and Western blot.RESULTS There was statistically significant difference in the cytostasis between CoFG treated group and the non- CoFG treated group.CoFG could down-regulate the expression of mRNA and protein of MMP-2 and Bcl-2 in treated CAL27 cells of OSCC,and CoFG could up-regulate the expression of mRNA and protein of MMP-9 and Bax in treated CAL27 cells of OSCC.CONCLUSION CoFG is an important traditional Chinese medicine which shows potential therapeutic effect on OSCC.
