摘要
目的:观察干扰素γ对舌鳞癌细胞增殖的影响。方法:采用不同浓度的干扰素γ对舌鳞癌细胞处理,观察肿瘤细胞的增殖情况;基因芯片分析干扰素γ处理后基因表达的变化,并采用实时定量PCR验证;western blot检测AKT信号通路的表达情况。结果:采用不同浓度的干扰素γ处理CAL27细胞后,肿瘤细胞的生长明显加快,肿瘤生长72、96h后,细胞数量有显著性差异(P<0.05)。基因芯片结果显示PIK3C2A的表达提高2.5倍以上。western blot结果证实干扰素γ处理细胞后,AKT信号通路被激活。结论:干扰素γ处理舌鳞癌细胞后能加快肿瘤细胞的生长,激活AKT信号通路,其不适用直接对舌鳞癌进行治疗。
Objective:To observe the effect of Interferon γ(IFN-γ) in regulating the growth of oral squamous cell carcinoma(OSCC)cell lines.Methods:Different doses of IFN-γ were added to CAL27 cells.Cell number was counted at the set time points.Gene profile chip was used to detect the expression change of various genes.Realtime PCR was applied to test the results.Expression change of components in AKT signaling was detected by Western blot.Results:The growth of CAL27 was accelerated dramatically with the treatment of different dose.After 72 and 96 hours,the number of tumor cells demonstrated significant difference(P0.05).The result of gene chip revealed over 2.5 fold increase of PIK3C2A expression,which was confirmed by realtime PCR.Western blot proved the activation of AKT signaling.Conclusion:IFN-γ could accelerate the growth of OSCC cells and activate AKT signaling.Direct treatment of IFN-γ against OSCC was not suggested.
